摘要
目的:探讨尖叶假龙胆醇提物(EEGA,ethanol extract from Gentianella acuta)含药血清对人肺癌A549细胞株诱导凋亡和影响细胞周期的作用。方法:采用四甲基偶氮盐(MTT)比色法检测EEGA含药血清对A549细胞作用24 h、48 h、72 h的增殖抑制率,使用流式细胞仪观察EEGA含药血清处理A549细胞48 h后,细胞凋亡率和细胞周期阻滞率变化的检测。结果:MTT显示EEGA低、中、高剂量含药血清都能抑制A549细胞增殖(P<0.05),其抑制率与作用时间相关。低、中、高剂量含药血清处理A549细胞48 h后凋亡率显著高于空白组(P<0.05),同时低、中、高剂量含药血清处理A549细胞48 h后,G0/G1期细胞明显较空白组增加(P<0.01),提示EEGA含药血清可促进细胞凋亡,并且将细胞生长周期阻滞于G0/G1期。结论:尖叶假龙胆对人肺癌A549细胞有抑制作用,机制可能与抑制细胞生长、阻滞细胞生长周期及诱导细胞凋亡有关。
Objective: To investigate the effect of medicated serum of ethanol extract from Gentianella acuta( EEGA) on growth inhibition and induced apoptosis of human Lung cancer A549 cells. Methods: The MTT method was used to detect the cell growth inhibition rate after 24 h,48 h and 72 h,and flow cytometry instrument was used to observe the changes of apoptosis and cell cycle arrest of A549 cells after serum containing EEGA. Results: MTT showed that serum containing EEGA with low,medium,high doses could inhibit the proliferation of A549 cells( P 〈0. 05),which was related to the function time. According to low,medium and high dose drug- containing serum treatment after 48 h A549 cell apoptosis rate was significantly higher than that in the blank group( P〈 0. 05). At the same time according to low,medium and high dose of medicated serum processing A549 cells after 48 h,G0 / G1 phase cell obviously increased than that in the blank group( P 〈0. 01). It showed Gentianella acuta could promote cell apoptosis,and arrested in the cell cycle in G0 / G1 phase. Conclusion: Gentianella acuta has an inhibitory effect on the human lung cancer A549 cells,and the mechanism may be related to inhibiting the growth of cells,blocking the cell cycle and inducing apoptosis.
