ERIC-PCR技术分析葛根芩连汤对抗生素相关性腹泻模型肠道菌群结构的影响 被引量:9

Analysis of the effect of Gegenqinlian decoction on the intestinal microflora structure of an AAD model using ERIC- PCR technique

摘要 为了探讨葛根芩连汤对抗生素相关性腹泻(AAD)模型肠道菌群的微生态的调理作用,试验复制AAD模型,将试验动物随机分为对照组、模型组和用药组,用药组用葛根芩连汤进行治疗,于给药7天和14天分别处死巴马小型猪并收集不同肠段的内容物(包括十二指肠、空肠、回肠、盲肠和结肠)作为检测样品,采用ERIC-PCR技术对动物模型的十二指肠、空肠、回肠、盲肠和结肠中细菌菌群的结构多样性进行检测。结果表明:对照组巴马小型猪肠道菌群结构相对稳定,ERIC-PCR条带数量以盲肠最多,其次是结肠、十二指肠、空肠和回肠;模型组巴马小型猪各肠段ERIC-PCR扩增条带数相比较于对照组明显减少;用药组巴马小型猪各肠段ERIC-PCR扩增条带数随着用药时间的延长呈现正常的趋势。说明葛根芩连汤具有调整AAD导致的菌群紊乱的功能。 To investigate the adjusting effects of Gegenqinlian decoction on the micmecology of intestinal bacterial flora in an antibiotic - associ- ated diarrhea (AAD) model, the AAD model was replicated in the test. Experimental animals were randomly divided into control group, model group and administration group, and the administration group was treated with Gegenqinlian decoction. Bama miniature pigs were sacrificed at 7 d and 14 d after administration, respectively; and the contents from different intestinal segments of animal models, including duodenum, jejunum, ileum, cecum, and colon, were used as test samples. ERIC -PCR technique was used to detect the diversity of the bacterial flora structure in the duodenum, jejunum, ileum, cecum, and colon of animal models. The results showed that the intestinal bacterial flora structure in Bama miniature pigs from the control group was relatively stable, and the number of ERIC - PCR bands was maximum in the cecum, followed by colon, duodenum, jejunum and ileum; the number of ERIC - PCR bands in each intestinal segment of the Bama miniature pigs from the model group were significantly reduced compared with the control group ; and the number of ERIC - PCR bands in each intestinal segment of the Bama miniature pigs from the administration group exhibited normal trends with the elongation of administration time. The results indicate that Gegenqinlian decoction has the function to adjust flora disorder caused by AAD.
