FST基因在槟榔江水牛和德宏水牛中的遗传变异分析

摘要 为了研究卵泡抑素(FST)基因在槟榔江水牛和德宏水牛中的特征,试验采集43头槟榔江水牛和48头德宏水牛的血样,利用牛基因组报道的牛FST基因序列设计引物,采用PCR扩增和序列测定等分析技术,研究了这两种水牛的FST基因多态性、基因频率和基因型频率,构建了单倍型和单倍型亲缘关系聚类图,分析了杂合度、遗传分化系数等群体遗传特征。结果表明:FST基因在槟榔江水牛及德宏水牛中均有高度的多态性,在FST基因上共检测到4个单核苷酸多态性(SNPs)位点,从水牛单倍型间的遗传距离和单倍型聚类图的结构来看,槟榔江水牛与沼泽型德宏水牛亲缘关系较远。从FST核基因上为槟榔江水牛属于河流型水牛起源提供了分子遗传学依据。 To study the characteristics of follistatin (FST) gene in Binglangjiang buffaloes and Dehong buffaloes, the blood samples from 43 Binglangjiang buffaloes and 48 Dehong buffaloes were collected in the test. The primers were designed using the FST gene sequences reported by bovine genome, and the FST polymorphisms, gene frequencies and genotype frequencies of these two kinds of buffalos were studied using PCR amplification and sequencing and other analysis techniques. The haplotypes and the cluster diagram of haplotype phylogenetic relationships were constructed, and the heterozygosity, genetic differentiation coefficients and other population genetic characteristics were analyzed. The re- sults showed that the FST gene was highly polymorphic in Binglangjiang buffaloes and Dehong buffaloes, and four sites of single nucleotide poly- morphisms (SNPs) were detected in the FST gene. Binglangjiang buffaloes had a more distant relationship with Dehong buffaloes from the view of the genetic distance between Buffalo haplotypes and the structure of haplotype cluster diagram. The result provides a molecular genetic evi- dence for the origin of Binglangjiang buffalo belonged to river buffalo based on FST gene.
