摘要
目的探讨热休克蛋白70(HSP70)-Hom基因多态性与广西巴马人群长寿家族的相关性。方法于2010年7月,选取广西巴马县长寿人群后代101例(22~83岁)为有长寿家族史组,南丹县无长寿家族史的健康人群176例(20~82岁)为无长寿家族史组。对研究对象进行问卷调查和体格检查,采用TaqMan探针实时荧光定量PCR技术对HSP70-Hom基因rs2227956位点进行基因分型。结果有长寿家族史组和无长寿家族史组人群HSP70-Hom基因rs2227956位点基因型和等位基因频率分布差异具有统计学意义(P〈0.05)。经多因素logistic回归分析后,TT、CT基因型携带者(ORTT=5.161,95%CI:1.300~20.489;ORCT=2.245,95%CI:1.032~4.883)和高密度脂蛋白胆固醇升高者(OR=6.924,95%CI:2.484~19.301)有长寿家族史的比例较高,体质指数偏高者有长寿家族史的比例较低(OR=0.271,95%CI:0.092~0.796)。结论本研究初步认为有家族性长寿史者可能携带HSP70-Hom基因rs2227956位点的TT、CT基因型,且高密度脂蛋白胆固醇较高,体质指数偏低。
Objective To explore the relationship between heat shock protein 70(HSP70)-Hom polymorphism and people with familial longevity in Bama county of Guangxi. Methods A total of 101 descendants of longevity people, aged 22-83, from Bama county as the familial longevity history group and 176 healthy people aged 22-82 without familial longevity history from Nandan area as the non-familial longevity history group were selected in July 2010. All cases were investigated through the questionnaire and physical examination. Genotyping of HSP70-Hom(rs2227956) was performed by Taq Man real-time quantitative PCR. Results The differences in genotype and allele frequency of HSP70-Hom(rs2227956) distributions between the familial longevity history group and the non-familial longevity history group were significant(P0.05). The percentage of familial longevity history in TT/CT genotype carriers(ORTT=5.161,95% CI:1.300-20.489;ORCT=2.245,95% CI:1.032-4.883)and high density lipoprotein cholesterol increasing(OR =6.924,95% CI:2.484-19.301) were high. The percentage of familial longevity history in body mass index increasing was low(OR =0.271,95% CI:0.092-0.796). Conclusion This preliminary study shows that the people with familial longevity history may not only carry HSP70-Hom gene(rs2227956) TT/CT genotype,but also have higher level of high density lipoprotein cholesterol and have low body mass index.
