摘要
目的:观察补阳还五汤对局灶性脑缺血损伤模型大鼠微循环血流及相关因子的影响。方法:实验分为假手术组,模型对照组,阿托伐他汀组(10 mg·kg^(-1)),补阳还五汤低、中、高剂量组(5,10,20 g·kg^(-1));采用大脑中动脉栓塞法复制MCAO脑缺血再灌注大鼠模型,参照Longa法进行行为障碍评分;MoorFLPI散斑全帧实时扫描成像系统测定各大鼠尾部微循环血流;ELISA法检测血清TXB2、6-keto-PGF1α水平。结果:各剂量补阳还五汤均能显著降低模型大鼠神经行为学评分(P<0.05),增加尾部微血流量(P<0.05,P<0.01),20 g·kg^(-1)补阳还五汤能显著增加6-keto-PGF1α水平,降低TXB2/6-ketoPGF1α比例。结论:补阳还五汤对MCAO大鼠脑缺血损伤具有改善作用,该作用与改善微循环血流,调节TXB2/6-keto-PGF1α比例有关。
OBJECTIVE To observe effects of Buyang Huanwu decoction(BYHWD)on blood circulation and levels of correlation factors in rats with MCAO.METHODS SD rats were randomly divided into 6 groups:normal control group,model control group,positive control group(atorvastatin calcium capsules 10 mg·kg^-1)and BYHWD groups at doses of 5,10 and 20 g·kg^-1.MCAO model rats were established by middle cerebral artery occlusion.Tail microcirculation of MCAO rats was measured by Moor FLPI speckle imaging system,and contents of TXB2 and 6-keto-PGF1αwere measured by ELISA in serum.RESULTS BYHWD decreased behavioral scores(P〈0.05),increased flux of tail microcirculation effectively(P〈0.05,P〈0.01).BYHWD20 g·kg^-1 increased 6-keto-PGF1αcontent in serum and decreased ratio of TXB2/6-keto-PGF1αin rats with MCAO.CONCLUSION BYHWD can produce protective effects against cerebral ischemic injury probably by ameliorating microcirculation and regulating TXB2/6-keto-PGF1αratio.
