电针不同穴位对分娩大鼠行为学及中枢DA mRNA与蛋白表达的影响 被引量:1

Electric acupuncture acupoints on childbirth behavior of rats and the impact of central DA mRNA and protein expression

摘要 目的探讨不同穴位电针对分娩大鼠行为学及中枢DA、Drd2 mRNA与蛋白表达的影响。方法 120例孕鼠随机分为空白对照组、电针三阴交穴、合谷穴、合谷加三阴交、血海穴等不同穴位组及药物组。热水甩尾测痛观察电针穴位对大鼠行为学的影响、Real-Time PCR及Western blot检测大鼠中枢DA、Drd2 mRNA与蛋白表达。结果电针对分娩大鼠行为学影响显示,治疗前大鼠痛阈,组间差别(F=0.736,P=0.598>0.05)无统计学意义,治疗后,组间差别明显(F=216.361,P<0.01)有统计学意义,不同穴位依次为三阴交穴>合谷穴>合谷+三阴交>血海>不同程度提高大鼠痛阈;大脑灰质DA、Drd2 mRNA与蛋白表达水平,组间差异显著,有统计学意义(P<0.01),与空白组相比,电针各穴位组、药物组DA、Drd2 mRNA与蛋白表达水平明显降低,依次为空白组<血海组/合谷加三阴交组/合谷组<三阴交组<药物组;而脊髓DA、Drd2 mRNA与蛋白表达水平比较,组间差异不明显,无统计学意义(P>0.05)。结论电针穴位有镇痛作用;不同穴位有不同程度提高大鼠痛阈的作用,依次为三阴交穴>合谷穴>合谷+三阴交>血海>;不同穴位产生不同针效与电针由低至高依次为空白组<血海组/合谷加三阴交组/合谷组<三阴交组<药物组不同程度降低大脑灰质DA、Drd2 mRNA与蛋白表达水平呈负相关。揭示大脑灰质区为电针穴位DA镇痛作用机制的靶点。 Objective To discuss childbirth behavior of rats in different meridians electricity and central DA,Drd2 mRNA and protein expression. Methods Including 120 cases of pregnant rats were randomly divided into blank control group,three Yin cave,valley cave,li4 plus three vaginal sex,blood point acupuncture group and drug group. Hot pain spin test to observe the influence of electric acupuncture acupoints on rat ethology,Real- Time PCR and Western blot detection rat central DA,Drd2 mRNA and protein expression. Results Electricity for delivery in the rat behavior influence,according to the pain threshold of rats before treatment,differences between groups( F = 0. 736,P = 0. 736 〉 0. 05) with no statistical significance,after treatment,the difference between groups( F = 216. 361,P 〈 0. 01) was statistically significant,different meridians followed by three vaginal intercourse hole hole 〉 close valley valley + vaginal intercourse three 〉 blood 〉 different levels increase the pain threshold of rats; Brain gray matter DA,Drd2 mRNA and protein expression level,the significant difference between groups was statistically significant( P 〈 0. 01),compared with the blank group,electric acupuncture acupoints group,the drug group DA,Drd2 mRNA and protein expression level decreased obviously,followed by a blank 〈 blood group / li4 plus three Yin / group / li4 〈 three vaginal intercourse group medicine group; And spinal cord DA,Drd2 mRNA and protein expression level,no obvious difference between groups,no statistical significance( P 〉 0. 05). Conclusion Electric acupuncture meridians have analgesic action,Different acupoints have different degree of increase the pain threshold of rats,followed by three vaginal intercourse hole hole 〉 close valley valley + vaginal intercourse three 〉 blood 〉; Different acupuncture needle produce different effect with acusector from low to high followed by a blank 〈 blood group / li4 plus three Yin / group / li4 〈 〈 three vaginal intercourse group drug group reduce brain gray matter DA,different level Drd2 mRNA and protein expression level was negatively correlated,reveal the area of gray matter for electric acupuncture acupoints DA analgesia mechanism of targets.
