婴幼儿配方乳粉中叶黄素含量测定的研究

Determination of lutein in infant formula milk powder

目的：研究利用高效液相色谱（HPLC）法测定婴幼儿配方乳粉中的叶黄素含量。方法：乳粉样品经水溶解,用含0.1%BHT的丙酮提取所含叶黄素,饱和氯化钠分离丙酮,上清液采用C18色谱柱分离,在445 nm波长下检测。结果：叶黄素在（0.02～0.8）mg/L范围内线性关系良好,相关系数为0.997,以3倍信噪比（S/N〉3）计方法的检出限为0.016 mg/L。在100、200、300μg/100 g的添加水平下,叶黄素的平均回收率为98.2%～103.9%,RSD为1.25%～2.53%。应用该方法检测了7种市售婴幼儿配方乳粉中的叶黄素含量,结果表明方法简便、准确。结论：此方法适合婴幼儿配方乳粉中叶黄素含量的日常检测。

Objective： Determination of lutein in infant milk powder by HPLC. Method： Milk powder was dissolved in water, then lutein were extracted with acetone containing 0.1% BHT. Saturated sodium chloride was used to separate acetone from water. The supernatant was determined with C18 column and detected at 445 nm. Results： The results showed that the linear range was （0.02-0.8） mg/L （r=0.997）, and the limit of detection was 0.016 mg/L. The mean recoveries of lutein were 98.2%-103.9% with the relative standard deviations of 1.25%-2.53%. The method was successfully applied to the analysis of lutein in 7 infant formula milk powder. Conclusion： The method was simple, precise and suitable for the daily determination of lutein in infant formula milk powder.
