摘要
目的探讨半乳凝素-3(galectin-3,gal-3)在小鼠胚胎着床子宫内膜中的表达及其与子宫内膜容受性标志分子整合素(integrin)β3的共表达定位。方法分别采用real-time PCR、Western blot和免疫荧光技术检测比较小鼠着床位点及非着床部位gal-3 mRNA和蛋白的表达及其与配体整合素β3、纤连蛋白(fibronectin,FN)的免疫荧光共定位表达。结果小鼠着床处gal-3 mRNA和蛋白表达高于非着床部位(P<0.01),其在着床位点主要与整合素β3共定位表达,而与FN的共定位极少。结论 gal-3在着床位点处高表达提示其在胚胎与子宫内膜着床的过程中扮演重要角色,且在着床处gal-3主要通过与整合素β3共表达参与胚胎着床。
Objective To explore the expression of galectin-3(gal-3)in mouse′s implantation site of endometrium and the co-localization with integrinβ3,which is the marker molecular molecular for the receptivity of endometrium. Methods Gal-3 mRNA and protein expressions were detected by realtime PCR and Western blot analysis.Immunofluorescence was used to detect the co-locatization of gal-3 and its ligands of integrinβ3 and fibronectin(FN). Results Compared with inter-implantation site,gal-3 mRNA and protein expressions in mouse implantation site of endometrium were significantly increased(P〈0.01).Moreover,the immunofluorescence intensity of the co-localization of integrinβ3and gal-3 was stronger in implantation site than that in inter-implantation site,while the co-localization of FN and gal-3 was very weak. Conclusions Gal-3 expresses highly in implantation site,and coexpresses with integrinβ3 at maternal-fetal interface.It indicates that gal-3 plays an important role in the attachment between embryo and endometrium.
