摘要
通过PCV2病毒感染3D4/21猪肺泡巨噬细胞后炎症相关细胞因子mRNA转录水平变化,探讨PCV2感染后可能的致病途径。通过PCV2病毒体外接种3D4/21细胞,感染不同时间后,收集样品,采用荧光定量PCR方法检测细胞因子IL-1β、IL-8及IL-18mRNA转录水平的变化。结果显示PCV2对3D4/21细胞的IL-1β、IL-8转录主要起抑制作用,对IL-8转录起促进作用。推测其在体内的综合效应导致感染早期机体抗病毒能力降低,引起机体免疫抑制,影响机体特异性免疫应答的正常运转,为PCVD的发病创造了条件。
In this study,we investigated the pathogenic pathways after PCV2 infection,by detecting the mRNA transcriptional profiles of inflammation-associated cytokines of porcine alveolar macrophages 3D4 /21 stimulated by PCV2. PCV2 strain inoculated with 3D /21 cells in vitro,and then collected samples in different infectious time. Last,the mRNA transcriptional level of cytokines IL-1β,IL-8 and IL-18 were detected by real-time PCR. The results showed that when PCV2 strain inoculated with 3D /21 cells in vitro,the mRNA transcription of IL-1β,IL-8 were inhibited,while the IL-18 were promoted. It was speculated that the combined effects resulted in the decrease of antiviral ability in early infectious stage in vivo,then caused immune suppression of the body to affect the normal function of specific immune response,and create conditions for the onset of PCVD.
