摘要
为了揭示脂肪型的藏猪和瘦肉型的杜洛克猪胰岛素样生长因子2(IGF-2)基因的表达差异,采用QRT-PCR方法检测了两猪种背最长肌中IGF-2基因在180日龄的表达差异,并分析其与肌纤维面积、肌内脂肪(IMF)含量的相关性。结果表明,180日龄藏猪IGF-2 m RNA表达量显著低于杜洛克猪(P<0.05)。相关性分析结果显示,IGF-2 m RNA表达量与肌纤维面积呈显著正相关(P<0.05),与IMF含量呈显著负相关(P<0.05)。以上结果初步揭示了两猪种在180日龄IGF-2基因表达的品种差异,为深入研究肌纤维生长及IMF沉积的调控机制提供了基础数据。
To investigate the differential expression of Insulin-like growth factor 2(IGF-2) gene between Tibet(a fatty, indigenous, Chinese breed) and Duroc(a leaner, Western breed) pigs. The expression differences of IGF-2gene in longissimus dorsi muscle were measured in two pig breeds at the age of 180 days by using QRT-PCR approach, their correlations with cross sectional area of muscle fiber(CSA) and intramuscular fat(IMF) content were also analyzed. The results showed that the expression level of IGF-2 m RNA in Tibet pigs was significantly lower than that of Duroc pigs at the age of 180 days(P〈0.05). The correlation analysis results showed that the expression level of IGF-2 m RNA had significant positive correlation with CSA(P〈0.05) and significant negative correlation with IMF content(P〈0.05). These results suggested that the m RNA level of IGF-2 gene in longissimus dorsi muscle are different in Tibet and Duroc pigs at the age of 180 days, which provided some data for further study on elucidating the molecular mechanism of growth of muscle fibers and IMF deposition.
