摘要
目的:研究石菖蒲不同极性部位高效液相图谱与抗抑郁效应的关联性。方法:采用系统溶剂法制备石菖蒲不同极性部位。采用梯度洗脱法建立石菖蒲原药材及不同极性部位的高效液相特征图谱,并确认图谱中特征峰及对应特征峰之间的峰面积数据关系。运用小鼠悬尾试验、强迫游泳试验等小鼠行为学实验,观察口服石菖蒲及不同极性部位抗抑郁效应。结果:石菖蒲环己烷、氯仿部分能显著缩短小鼠悬尾及游泳不动时间。石菖蒲抗抑郁效应与特征峰具有关联性。结论:石菖蒲具有抗抑郁作用,其主要抗抑郁效应来源于于环己烷及氯仿部位。
Objective: To explore the correlation of different polarity fractions of Rhizoma Acori Tatarinowii and thier antidepressant effects based on HPLC chromatogram. Methods: Different polarity fractions were prepared by the systematic solvent extraction method. HPLC characteristic chromatograms of crude drugs and different polarity fractions of Rhizoma Acori Tatarinowii were established with the gradient elution method; data relationship between characteristic peaks in chromatograms and corresponding peaks area were confirmed. The antidepression effects of Rhizoma Acori Tatarinowii and its different polarity fractions were evaluated with the behavior tests including the tails suspension test(TST) and forced swimming test(FST). Results: The cyclohexane and chloroform fraction obviously decreased the duration of immobility in the TST and FST. There was a correlation between the antidepressant effect of Rhizoma Acori Tatarinowii and its characteristic peak on chromatogram. Conclusion: Rhizoma Acori Tatarinowii performs an antidepressant effect, which is mainly derived from the cyclohexane and chloroform fraction. HPLC characteristic chromatography and data relationship between relative peaks could be used to judge the antidepressant effects of Rhizoma Acori Tatarinowii, and give a reference in the research and development in the antidepressant effects of Rhizoma Acori Tatarinowii.
