摘要
以中透艺春兰根状茎为材料,研究培养基和外源激素对根状茎增殖、分化的影响,同时探讨光照及不同根状茎材料对艺向稳定的影响。结果表明:3/4MS培养最适宜根状茎的增殖;TDZ 0.5 mg/L+6-BA 1.0 mg/L和6-BA 1.0 mg/L+NAA 0.3 mg/L的组合对根状茎的增殖效果较好;6-BA 1.0 mg/L+IBA 1.0 mg/L的组合最有利于根状茎的分化,芽分化率达到97.7%。2 000 lx的光照强度最有利于中透艺春兰的根状茎的出苗率和出艺率;选取整条为淡黄色的根状茎作为材料,能较好地保证叶艺的走向。
To establish the effects of different media and plant growth regulator treatments on proliferation and differentiation of rhizome, together with effects of different light radiation and rhizome material on directional art stability ,rhizome of Cymbidium goeringii "Zhung tou yi" were chose as material.The results showed that 3/4MS was the best basic media for proliferation of rhizome, and the best proliferation effect was found by adding TDZ 0.5 mg/L+6-BA 1.0 mg/L or 6-BA 1.0 mg/L+NAA 0.3 mg/L.The combination of 6-BA 1.0 mg/L+IBA 1.0 mg/L was good for differentiation of rhizome (97.7%).Light intensity of 2 000 lx was good for rate of plantlets and rate of leaf art.Moreover,the usage of material from pure light yellow rhizome ensured tendency of leaf art.
