华卟啉钠在SD大鼠体内代谢产物的质谱分析 被引量:1

摘要 目的考察灌胃给药华卟啉钠在大鼠体内的主要代谢产物和代谢途径。方法收集华卟啉钠灌胃后大鼠的血浆、尿液和粪便,采用Thermo Q Exactive Orbitrap四极杆-轨道阱高分辨质谱仪对大鼠尿、粪及血浆中的代谢产物进行研究,通过获得的精确分子量、二级碎片图谱,推断SD大鼠尿、粪及血浆中的代谢物,并绘制华卟啉钠在SD大鼠体内的代谢途径。结果在大鼠的尿、粪及血浆样品中共检测到10个代谢产物,主要为两卟啉之间的醚键断裂后的产物及其进一步加氢、N-甲基化和O-甲基化产物。结论华卟啉钠在大鼠体内以Ⅰ相代谢产物为主。 Objective To investigate the in vivo metabolism of sinoporphyrin sodium( DVDMS) in SD rats.Methods The blood,urine and feces samples were collected from the rats after oral administration of DVDMS and analyzed by a hybrid quadrupole-orbitrap mass spectrometer combining high-performance quadrupole precursor selection with high-resolution,accurate-mass to obtain the accurate molecular mass of parent ions and product ions.Metabolites were identified based on the accurate mass,fragmentation patterns,and multiple-stage mass data( MSn) so that the metabolic pathway of DVDMS in SD rats was postulated. Results A total of 10 metabolites in the blood,urine and feces of SD rats were detected and identified,mainly as the cleavage products of ether bond and their further hydrogenation,N-methylation and O-methylation products. Conclusion The main metabolic pathway of DVDMS in rats is phase Ⅰ,including ether cleavage,hydrogenation,N-methylation and O-methylation.
