猪FcγRⅢ双抗体夹心ELISA方法的建立及其在PRRSV感染猪中的初步应用

摘要 利用自行制备的FcγRⅢ多克隆抗体作为捕获抗体,鼠源抗FcγRⅢ特异性单克隆抗体作为示踪抗体,辣根过氧化物酶标记羊抗鼠Ig G作为检测系统,建立了检测可溶性FcγRⅢ(s FcγRⅢ)水平的双抗体夹心ELISA方法。利用建立的该ELISA方法对感染猪繁殖与呼吸障碍综合征病毒(PRRSV)血清进行检测,结果显示,猪感染PRRSV后,血清中s FcγRⅢ的水平明显增高,因此s FcγRⅢ的水平可能作为PRRSV感染的预警标志。该研究结果为进一步阐释FcγRⅢ在PRRSV感染中发挥的作用奠定了基础。 A double antibody sandwich ELISA was developed using rabbit anti-FcγRⅢ polyclonal antibody as the capture an-tibody,mouse anti-FcγRⅢ monoclonal antibody as the trace antibody and HRP-conjugated goat anti-mouse Ig G as the detectionsystem.The sandwich ELISA was used to detect porcine FcγRⅢ levels in serum from pigs infected with porcine reproductive andrespiratory syndrome virus(PRRSV).Our the results revealed that s FcγRⅢ levels in serum were significantly increased in pigs in-fected with PRRSV,suggesting that s FcγRⅢ may be referred as early warning of PRRSV infection.These results have laid a foun-dation for futher study on interpretation of FcγRⅢ in PRRSV infection.
