内质网应激通路在脑泰方提取物保护局灶性缺血大鼠海马神经元中的作用 被引量:5

摘要 目的:缺血性脑卒中是发病率越来越高的一种临床疾病,但是有关它的治疗还缺乏十分有效的手段,此实验的目的是通过探索中药复方(脑泰方)提取物作用于脑缺血的可能机制从而为临床提供有针对性的治疗方案。方法:自备中药复方益气活血方(脑泰方)连续灌胃7 d,在体制备大鼠大脑中动脉栓塞模型(MCAO),观察术后大鼠的神经行为,同时使用TTC染色观察神经元凋亡情况,RT-q PCR观察内质网应激通路相关蛋白的表达。结果:神经行为学评分反应出脑泰方(NTF)提取物可改善大鼠脑缺血后的神经行为;TTC染色显示脑泰方可明显增加MCAO后海马CA1区神经元的存活。RT-q PCR显示脑泰方能明显促进GRP78基因的表达,同时抑制CHOP基因的表达。结论:内质网应激参与了缺血导致的大鼠大脑海马神经元的损伤。益气活血法对脑缺血引发的内质网应激具有一定的干预作用,尤其是在脑缺血的早期。 Objective: To observe the possible mechanisms of the extraction of Naotai Fomula( NTF) on the regional cerebral ischemia so as to provide treatment to the more and more common disease-cerebral arterial thrombosis. Methods: Self-made Naotai Fomula were given to SD rats by gavage administration for 7 days before the occlusion of the middle cerebral artery( MCAO) operation. After the operation,the TTC staining was used to show the apoptosis of the hippocampal neurons and RT-PCR was used to show gene expression of GRP78 and CHOP of the ischemia cerebrum of rats. Results: Firstly,the extraction of NTF could improve the neuro behavior score and at the same time decrease the death rate of the hippocampal neuron after MCAO operation. Secondly,the extraction of NTF elevated the expression of GRP78 and decreased the expression of CHOP. Conclusion: Firstly,ER Stress is involved in the hippocampal neuronal injury caused by the ischemia in the rats. Secondly,the extraction of NTF can regulate ERS especially in the early stage after acute ischemia of the brain in rats.
