马铃薯提取液对人卵巢癌细胞株HO-8910增殖及磷酸化ERK1/2蛋白表达的影响 被引量:1

摘要 目的观察马铃薯提取液对人卵巢癌细胞增殖及磷酸化细胞外调节蛋白激酶1/2(ERK1/2)蛋白表达的影响。方法培养人卵巢癌细胞株HO-8910,待98%细胞生长状态良好、长满96孔细胞培养板时开始实验。将细胞分为实验1、2、3、4组及对照组。实验1、2、3、4组分别加入13、25、50、100μL/m L的马铃薯提取液,对照组不添加。各组分别于培养24、48、72 h采用MTT法检测光密度值(OD值),计算细胞增殖抑制率。收集各组细胞,分别于培养0、10、20、40 min采用Western blotting法检测磷酸化ERK1/2蛋白。结果培养24 h时实验4组细胞增殖抑制率高于实验1、2、3组(P均<0.05);培养48 h时实验3、4组细胞增殖抑制率高于实验1组,实验4组细胞增殖抑制率高于实验2组(P均<0.05);培养72 h时实验2、3、4组细胞增殖抑制率高于实验1组(P均<0.05)。培养20、40 min时实验3组细胞中磷酸化ERK1/2蛋白相对表达量低于同组培养0、10 min时(P均<0.05);培养20、40 min时实验3组细胞中磷酸化ERK1/2蛋白相对表达量低于同时点实验1、2、4组(P均<0.05)。结论马铃薯提取液可抑制人卵巢癌细胞增殖,并下调细胞内磷酸化ERK1/2蛋白表达。
