摘要
目的:观察填精补脑方对老年痴呆大鼠认知功能障碍的治疗作用。方法:健康成年SD大鼠随机分为对照组、模型组、脑复康组和填精补脑方组,每组10只。双侧海马注射Aβ25-35构建老年痴呆大鼠模型,各组给予相应药物灌胃治疗20 d。用药结束后,测定大鼠的学习记忆能力并检测海马CA1区长时程增强(LTP)。结果:与模型组相比,用药组大鼠学习和记忆能力均明显增强(P<0.01),海马LTP幅度显著升高(P<0.01)。结论:填精补脑方能够改善老年痴呆大鼠认知功能障碍,其作用机制可能与提高大鼠海马CA1区的LTP,明显改善突触的可塑性有关。
Objective: To observe the therapeutic effect of Tianjing Bunao Prescription in treating cognitive dysfunction in the Alzheimer disease rats. Methods:Healthy adult SD rats were randomly divided into 4 groups: the control group, the model group, Piracetam group and Tianjing Bunao Prescription group, with 10 rats in each group. The model of Alzheimer disease rats was established with microinjection Aβ25-35 on bilateral hippocampus. All rats were treated with corresponding drugs respectively, which lasted 20 days. Then, the learning and memory ability of rats were examined. Long term potentiation(LTP) in hippocampal CA1 region was detected. Results: The learning and memory ability and the LTP in CA1 area of hippocampus of rats in Tianjing Bunao Prescription group improved significantly compared with that of the model group(P<0.01). Conclusion: Tianjing Bunao Prescription could improve the cognitive dysfunction of AD rats, of which the mechanism may be closely related to the enhancement of LTP in CA1 area of hippocampus and the significant improvement of synaptic plasticity.
