鳖甲煎口服液对肝纤维化大鼠ACE-AngⅡ-AT1R轴的影响 被引量:7

Effect of Biejiajian Oral Liquid on ACE-Ang Ⅱ-AT1R Axis in Liver Fibrosis Rats

摘要 [目的]研究鳖甲煎口服液(Biejiajian oral liquid,BOL)对酒精性肝纤维化大鼠肝组织中血管紧张素转换酶-血管紧张素Ⅱ-血管紧张素Ⅱ1型受体(angiotensin converting enzyme-angiotensinⅡ-angiotensinⅡtype 1 receptor,ACE-AngⅡ-AT1R)轴的影响。[方法]将65只雄性SD大鼠随机分为正常组、模型组、马来酸依那普利(enalapril malecate,Ena)组、BOL低、中、高剂量组,其中模型组15只,其余每组10只。除正常组外其余各组大鼠制备酒精性肝纤维化模型,造模结束后各组给予相应药物治疗4周。采用苏木精-伊红(hematoxylin-eosin,HE)染色观察各组大鼠的肝纤维化状况,酶联免疫法(enzyme linked immunosorbent assay,ELISA)检测肝组织中的AngⅡ的水平,以实时定量PCR(real-time polymerase chain reaction,RT-PCR)检测肝组织中ACE、AT1R的mRNA表达。[结果]BOL各剂量组和Ena组的肝纤维化程度减轻,模型组肝组织中的AngⅡ含量和ACE、AT1R mRNA表达高于正常组,差异有统计学意义(P<0.05);BOL各剂量组和Ena组可以显著降低肝组织中的AngⅡ含量和ACE、AT1R mRNA的表达。[结论]AngⅡ含量和ACE、AT1R mRNA表达升高可能是大鼠酒精性肝纤维化发生机制之一,而BOL可以通过下调三者的表达对大鼠酒精性肝纤维化的发展起到抑制作用。 [Objective]To study the effect of Biejiajian oral liquid(BOL) on ACE-AngⅡ-AT1 R axis in liver fibrosis rats. [Method]The liver fibrosis model was induced by alcohol-corn oil-parazole. After being duplicated successfully, every group was made i.g. BOL respectively for 4 weeks, and enalapril as positive control. The fibrosis condition was observed by hematoxylin-eosin(HE) staining, the level of Ang Ⅱ in liver homogenate was determined by Enzyme linked immunosorbent assay(ELISA), and real-time polymerase chain reaction(RT-PCR) was applied to measure the expression of ACE and AT1 R mRNA in liver homogenate. [Result] The degree of liver fibrosis of BOL group and enalapril group were lighter than that of the model group. The level of AngⅡ and the expression of ACE and AT1 R mRNA in liver homogenate significantly increased compared with the model group(P〈 0.05). BOL and enalapril could significantly reduce the level of AngⅡ and the expression of ACE and AT1 R mRNA. [Conclusion] Elevated expression of ACE-AngⅡ-AT1 R axis may be one of mechanisms of rat liver fibrosis. BOL can inhibit the development of rat liver fibrosis by reducing the expression of ACEAngⅡ-AT1 R axis.
