浸润性乳腺癌中DJ-1和PTEN的表达及临床意义研究 被引量:1

Expression of DJ-1 and PTEN in Invasive Breast Cancer and its Correlation with Relative Clinical Parameters and Prognosis

摘要 目的:探讨浸润性乳腺癌组织中DJ-1和抑癌基因(PTEN)的表达,分析其相关性及临床意义。方法:采用免疫组化(SP)法检测DJ-1、PTEN蛋白在150例浸润性乳腺癌和50例正常乳腺组织的表达,分析两者的相关性并探讨其与浸润性乳腺癌临床病理参数关系和对预后的影响。结果:浸润性乳腺癌DJ-1的阳性率61.3%显著高于乳腺正常组织8.0%(P<0.05),PTEN的阳性率20.7%显著低于乳腺正常组织96.0%(P<0.05)。DJ-1、PTEN蛋白在浸润性乳腺癌组织中的表达与淋巴结转移、组织学分化和TNM分期密切相关(P<0.05)。浸润性乳腺癌组织中DJ-1蛋白表达与PTEN蛋白表达呈负相关((P<0.05)。DJ-1阳性的乳腺癌组织Ki-67的阳性细胞数约33%与DJ-1阴性乳腺癌组织Ki-67的阳性细胞数约11%比较,差异有统计学意义(P<0.05)。在5年无瘤生存率和5年总生存率方面,DJ-1表达阴性者高于阳性者(P<0.05);PTEN表达阳性者高于阴性者(P<0.05)。结论:浸润性乳腺癌DJ-1表达增高,同时PTEN表达下降,统计学分析两者呈负相关。DJ-1高表达、PTEN低表达为浸润性乳腺癌患者预后的独立危险因素,参与调控浸润性乳腺癌的发生及预后。 Objective To investigate the association among the expression of DJ-1 and PTEN AR in invasive breast cancer tissues,and explore the correlation between the expression and its relative clinical parameters. Methods We collected 150 cases of invasive breast cancer and 50 cases of normal people to determin the expression of DJ-1 and PTEN by using immunohistochemical staining. Results The positive expression rates of DJ-1 and PTEN in invasive breast cancer were 61.3 %(92/150)and 20.7 %(31/150)respectively.In contrast,the positive expression of DJ-1 and PTEN in normal tissues was only 8.0 %(4/50)and 96.0 %(48/50). Compare to normal tissues,the positive expression rates of DJ-1 was significantly increased in invasive breast cancer(P 0.05), and PTEN positive expression in TNBC was significantly decreased(P 0.05). The expression of DJ-1 and PTEN in invasive breast cancer were significantly associated with lymph node metastasis,differentiation and TMN stages(P 0.05). In 150 invasive breast cancer tissues,DJ-1 and PTEN expression was negatively correlated(P 0.05). DJ-1 and PTEN were strong independent prognostic factors for five years of disease-free surviva1 rate of invasive breast cancer patients(P 0.05). Conclusions DJ-1 and PTEN may contribute to the development of invasive breast cancer and are valuable prognostic factors for invasive breast cancer patients. DJ-1 may inhibit PTEN expression. The cooperation of the two proteins may be related to the prognosis of invasive breast cancer.
