脂联素在应激状态下对小鼠卵泡发育的影响 被引量:2

Effect of Adiponectin on Follicular Development Under Stress in Mice

摘要 目的:利用饥饿刺激探讨应激状态下脂联素缺失对小鼠卵泡发育的影响。方法:C57BL/6、脂联素半缺失(APN+/-)、脂联素全缺失(APN-/-)三种基因型小鼠以正常进食量的一半给予饥饿刺激,建立应激模型,30天后处死小鼠取卵巢,计数三种基因型小鼠各级卵泡数目。结果:1经过30天饥饿后饥饿组小鼠体重下降与初始体重比较有统计学意义(P<0.001),应激模型建立。2应激状态下三种基因型小鼠卵巢重量未见统计学差异(P>0.05)。3正常饮食组三种基因型小鼠各级卵泡比例未有统计学差异(P>0.05)。饥饿后C57BL/6小鼠原始卵泡比例为(47±2.966)%,APN+/-原始卵泡比例为(36.5±1.555)%(P<0.05),APN-/-原始卵泡比例为(36.8±2.200)%(P<0.05);C57BL/6闭锁卵泡比例为(12±1.225)%,APN+/-闭锁卵泡比例为(19.75±1.887)%(P<0.01),APN-/-闭锁卵泡比例为(20±0.8367)%(P<0.001),与野生型闭锁卵泡比例比较差异有统计学意义。结论:饥饿刺激下脂联素缺失小鼠原始卵泡消耗增加,而闭锁卵泡的发生增加,处于生长状态的卵泡减少,提示在应激状态下脂联素水平的下降可导致卵泡成熟的过程受阻。 Objective: To study the effect of adiponectin on the follicular development in mice under stress. Methods: C57B6/L,Adiponectin heterozygotic(APN+/-), Adiponectin homozygotic(APN-/-) were fed by half of the diet. The mice were killed after 30 days,and the number of follicles was counted and analyzed. Results: 1 After 30 days of starvation, the weight of the mice was decreased significantly(P〈0.001), and the model of stress were established. 2 The weight of ovary showed no difference between the three genetypes under stress(P〈0.05). 3 The three genotypes of normal diet showed no significant difference in the percentage of follicles(P〈0.05). The percentage of primordial follicles in the wild type mice, heterozygotic and homozygotic mice were(47±2.966)%,(36.5±1.555)% and(36.8±2.200)%, respectively, and the difference had statistical significance(P〈0.05). The percentages of atretic follicles in the wild type was(12 ±1.225)%, while the percentages of atretic follicles was(19.75 ±1.887)%(P〈0.01) in the adiponectin heterozygotic mice and(20.00±0.8367)%(P〈0.001) in the adiponectin KO mice, which was statistically significant compared with that of the wild type mice. Conclusions: Starvation can stimulate the consumption of the primordial follicles of adiponectin deficient mice,while the number of atretic follicles is increased, suggesting that the decrease of adiponectin levels under stress can lead to the obstruction of follicular maturation process.
