摘要
目的:探讨黄芪注射液对盲肠结扎穿孔(cecal ligation and puncture,CLP)致脓毒症大鼠胸腺细胞凋亡的影响及机制。方法:将大鼠随机分成正常对照组、模型组、黄芪高剂量组、黄芪中剂量组及黄芪低剂量组。采用CLP致脓毒症模型,应用TUNEL法及光镜、透射电镜观察CLP致脓毒症后36 h大鼠胸腺细胞的凋亡情况,并应用免疫组化法检测胸腺细胞Caspase-3和Bcl-2蛋白表达的变化。结果:CLP致脓毒症可导致大鼠胸腺细胞凋亡增多,电镜和光镜均显示其典型的形态学特征。CLP致腹腔感染后胸腺细胞Caspase-3蛋白表达增加,Bcl-2蛋白表达下降。黄芪注射液可减轻CLP致大鼠脓毒症后出现的胸腺细胞凋亡,减少胸腺细胞Caspase-3蛋白表达,同时提高胸腺细胞Bcl-2蛋白表达。结论:黄芪注射液可以减轻CLP致脓毒症大鼠胸腺细胞凋亡,其机制与减少胸腺细胞Caspase-3表达、提高Bcl-2蛋白表达有关。
Objective: To explorethe influence of Huangqi Injection on apoptosis of thymocytes in rats with sepsis and its mechanism. Methods: Sprague- Dawley( SD) rats were randomly divided into five groups,including normal control group,model group,high dose Huangqi group,medium dose Huangqigroup and low dose Huangqi group. The septic models were made from cecal ligation and puncture( CLP). The apoptosis of thymocytes was observed under electron microscope and light microscope,and apoptosis ratio of thymocytes was determined by TUNEL method. The expressions of Caspase- 3 and Bcl- 2 protein in thymocytes were determined by immunochemical method. Results: The apoptosis of thymocytes in sepsis rat resulted from CLP was significantly increased and the typical histopathology changes of apoptosis of thymocytes were observed under electron microscope and light microscope. The expression of Caspase- 3 protein on thymocytes was significantly increased and expression of Bcl- 2 protein was reduced. The changes were improved significantly in Huangqi Injection group,the expression of Bcl- 2 protein of thymocytes was also significantly increased and expression of Caspase- 3 protein was decreased in Huangqi Injection group. Conclusion: Huangqi Injection can significantly reduce the apoptosis of thymocytes in rats with sepsis resulted from CLP and that may be closely associated with the increasing the expression of Bcl- 2 protein and decreasing the expression of Caspase- 3 in thymocytes.
