氟马西尼对盐酸氯胺酮麻醉大鼠学习记忆功能的影响 被引量:3

摘要 目的 探讨氟马西尼对盐酸氯胺酮注射液麻醉后大鼠学习记忆的影响及其可能机制。方法 选择60只健康清洁级SD大鼠,随机分成对照组、氯胺酮组、氟马西尼组各20只。对照组:第1天腹腔注射生理盐水,第2~6天无药物处理。氯胺酮组:第1天腹腔注射盐酸氯胺酮(80 mg/kg),从第2开始每日腹腔注射生理盐水,持续至第6天。氟马西尼组:第1天腹腔注射盐酸氯胺酮(80 mg/kg),从第2天开始每日腹腔注射氟马西尼(4 mg/kg),持续至第6天。采用Morris水迷宫检测氟马西尼对盐酸氯胺酮所致学习记忆障碍的影响,同时采用酶联免疫吸附(ELISA)方法检测各组大鼠海马区脑源性神经营养因子(BDNF)和神经生长因子(NGF)的含量,并采用Western印迹检测各组大鼠海马组织c AMP反应元件结合蛋白(cREB)、磷酸化cREB(p-cREB)、细胞外信号调节蛋白激酶(ERK)、p-ERK的表达水平。结果 第2天和第3天时各组间逃避潜伏期均无显著差异;第4~6天,与对照组相比,氯胺酮组寻找平台的逃避潜伏期明显延长(P〈0.05)。与氯胺酮组相比,氟马西尼组的逃避潜伏期于第5~6天时缩短明显(P〈0.05)。氯胺酮组大鼠穿越原平台位置次数少于氟马西尼组(P〈0.05)。氟马西尼组大鼠穿越原平台位置次数少于对照组(P〈0.05)。与对照组相比,氯胺酮组大鼠海马匀浆中NGF、BDNF的含量下降,与氯胺酮组比较,氟马西尼组含量增加(均P〈0.05)。与对照组相比,氯胺酮组海马p-cREB、p-ERK的蛋白表达水平均显著降低,与氯胺酮组比较,氟马西尼组表达水平均显著增加(均P〈0.05)。结论 氟马西尼可能通过增加NGF和BDNF表达水平,激活ERK、cREB信号通路,从而改善盐酸氯胺酮所致的学习记忆障碍。
