葡萄糖调节蛋白78在妊娠期糖尿病孕妇胎盘滋养细胞中的表达

The expression of glucose regulated protein 78 in placenta trophocytes of pregnant women with gestational diabetes mellitus

目的探讨研究葡萄糖调节蛋白78在妊娠期糖尿病孕妇胎盘滋养细胞中的表达情况,了解其在妊娠糖尿病临床诊断及预后观察方面的临床意义。方法选取2014年10月至2015年10月收治的妊娠期糖尿病患者85例作为观察组,选取同时间段收治的健康妊娠期女性100例设为对照组,2组孕妇均因不同原因分娩实施剖宫产手术,检测所有入组对象葡萄糖调节蛋白78在胎盘滋养细胞中的表达情况。葡萄糖调节蛋白78 mRNA以实时荧光定量PCR检测,葡萄糖调节蛋白78蛋白以免疫组化二步法检测,分级按照细胞染色强度实施。结果葡萄糖调节蛋白78 mRNA方面:观察组(16.2±0.6)与对照组(6.9±0.4)比较,差异有统计学意义(P<0.05);葡萄糖调节蛋白78蛋白强阳性表达率方面:观察组(96.47%)与对照组(25.00%)比较,差异有统计学意义(P<0.05)。其中观察组为深棕色、浅棕色、黄色染色强阳性表达,对照组仅25例为浅棕色染色强阳性表达,其他均为黄色染色弱阳性表达。结论葡萄糖调节蛋白78在妊娠期糖尿病孕妇胎盘滋养细胞中为高表达情况,可能和妊娠期糖尿病的发病存在关联。

Objective To observe the expression of glucose regulated protein 78 in placenta trophocytes of pregnant women with gestational diabetes mellitus in order to investigate its significance in clinical diagnosis and prognosis evaluation of gestational diabetes mellitus.Methods Eighty-five patients who were admitted and treated in our hospital from October 2014 to October 2015 were served as observation group,besides,the 100 healthy gestational women were served as control group,the pregnant women in both groups underwent cesarean section due to different causes.The expression levels of glucose regulated protein 78 in placenta trophocytes were detected in both groups.The expression levels of glucose regulated protein 78 mRNA were detected by Real-time fluorescent quantitative PCR and expression levels of glucose regulated protein 78 protein were detected by immunohistochemisty two stage method.Results The expression levels of glucose regulated protein 78 mRNA in observation group（ 16.2 ± 0.6） were significantly higher than those（ 6.9 ± 0.4） in control group（ P〈0.05）.The expression levels of glucose regulated protein 78 protein in observation group（ 96.47%）） were significantly higher than those（ 25.00%） in control group（ P〈0.05）,in which the strong positive expression with dark brown,light brown,yellow staining was found in observation group,however,the strong positive expression with light brown was observed only in 25 patients of control group,and weakly positive expression with yellow staining was found in the other patients of control group.Conclusion The high-expressions of glucose regulated protein 78 exist in placenta trophocytes of pregnant women with gestational diabetes mellitus,which may be correlated to the pathogenesis of gestational diabetes mellitus.
