摘要 [目的]优化橡胶草再生和遗传转化体系。[方法]以橡胶草茎和叶为外植体,以1/2MS和MS为基本培养基,研究不同激素浓度配比、外植体类型和激素浓度对橡胶草不定芽诱导和生根的影响。[结果]橡胶草进行组织培养的最佳外植体类型为茎,诱导不定芽形成的最佳激素浓度配比为1.0 mg/L 6-BA和0.10 mg/L NAA。诱导不定芽生根的最佳激素浓度为0.10 mg/L NAA,生根率达100%,移栽成活率达95%以上。[结论]建立了橡胶草高效再生体系,对提高橡胶草组织培养的效率和转化率具有重要意义。 [Objective] The aim was to optimize Taraxacum kok-saghyz Rodin regeneration and genetic transformation system. [Method] With stems and leaves of T. kok-saghyz as explants,1 /2MS and MS as the basic medium,effects of different hormone concentrations and explant types on adventitious bud induction and rooting were studied.[Result] The results showed that the best explant for tissue culture was stem,the best hormone concentration ratio of adventitious bud induction was 1. 0 mg / L 6-BA and 0. 10 mg / L NAA,and the adventitions bud was healthy. The most suitable hormone concentration for adventitious bud rooting was 0. 10 mg / L NAA with 100% rooting rate and 95% survival rate. [Conclusion] The established high efficient regeneration system of T. kok-saghyz has significance on improving tissue culture efficiency and transformation ratio.
