玉屏风水提液对维甲酸诱导大鼠骨质疏松的预防作用 被引量:1

摘要 目的了解玉屏风水提液对维甲酸诱导大鼠骨质疏松的预防作用。方法雌性SD大鼠30只随机分成正常组、维甲酸组和玉屏风组,分别给予对照溶液、维甲酸70 mg/(kg·d)和维甲酸+玉屏风水提液5 m L/(kg·d)灌胃,连续28d;处死前第13、14天和3、4天分别皮下注射四环素、钙黄绿素进行荧光标记,处死后分别取股骨、胫骨测量骨密度、骨生物力学、骨形态计量学。结果与正常组对比,维甲酸组股骨重量、骨密度、骨生物力学参数均下降(P<0.01或0.05),胫骨松质骨总面积、骨小梁面积、骨小梁周长、标记周长百分数、骨形成率及皮质骨面积和骨外膜骨形成降低(P<0.01或0.05),而骨内膜骨形成增强(P<0.01)。与维甲酸组对比,玉屏风组骨生物力学参数增加(P<0.01或0.05),而其他参数差异无统计学意义(P>0.05)。结论玉屏风水提液可预防维甲酸所致大鼠骨生物力学降低,但对骨密度和形态计量学无明显影响。 Objective To investigate the preventive effect of Yupingfeng water extract on retinoic acid(RA)-induced osteoporosis in rats. Methods Thirty female SD rats were randomly divided into control, RA and Yupingfeng groups, of which received the intragastric administration of control solution, 70 mg/(kg·d) RA, and RA plus 5 mL/(kg·d) Yupingfeng water extract for 28 days, respectively. All rats were subcutaneously injected with tetracycline on days 13 and 14, and calcein on days 3 and 4 before sacrifice. Bone weight, density, biomechanics and histomorphometry were measured in the femur and tibia. ResultsCompared with control group, bone weight, density and biomechanic parameters of the femur were significantly decreased in RA group(P〈0.01 or 0.05); total area, trabecular area and perimeter, percent labeled perimeter, and bone formation rate of the cancellous tibia, as well as total area and periosteal formation of the cortical tibia were reduced(P〈0.01 or 0.05), while endosteal formation of the cortical tibia were increased(P〈0.01) in RA group. The remarkable improvement of bone biomechanic indexes(P〈0.01 or 0.05) but not other parameters were observed in Yupingfeng group compared with RA group. ConclusionYupingfeng water extract can prevent RA-induced biomechanical deterioration, but has no effect on bone density and histomorphometry in rats.
