摘要
目的探讨大鼠颅脑损伤后血浆、海马内皮素(ET-1)及降钙素基因相关肽(CGRP)水平的变化,以及三七总皂甙(PNS)对其影响。方法 80只Wister大鼠随机分为空白组、模型组、PNS低剂量组、PNS高剂量组各20只,采用Feeney法建立大鼠颅脑损伤模型,RIA法测定颅脑损伤72小时大鼠血浆及海马中ET-1、CGRP的含量,HE和Niss染色观察大鼠大脑及海马病理变化。结果与空白组比较,模型组大鼠血浆及海马中ET-1显著升高(P<0.05),CGRP明显下降(P<0.05);与模型组比较,PNS低、高剂量均能显著降低血浆及海马中ET-1水平(均P<0.05),升高CGRP含量(均P<0.05);高剂量组与低剂量组ET-1及CGRP水平比较均有显著性差异(均P<0.05),提示PNS能减轻脑损伤大鼠脑组织的细胞形态学变化,减少海马神经元的死亡数量。结论三七总皂甙可显著降低颅脑损伤大鼠血浆及海马中ET-1含量,升高CGRP水平,减轻颅脑损伤后继发性脑损害。
Objective To explore the change and the effect of Panax notoginseng(PNS)on the levels of endothelin-1(ET-1)and Calcitonin gene-related peptide(CGRP)in the plasma and hippocampus of rats with brain trauma.Method 80 Wister rats were randomly divided into blank group,model group,PNS low dose group and PNS high dose group.The brain trauma model was established by improved Feeney's method.ET-1and CGRP in the plasma and hippocampus of rats were determined by RIA.The pathological change of brain and hippocampus was observed with HE and Niss dyeing.Result Compared with that of the blank group,the levels of ET-1in the plasma and hippocampus of of model group significantly increased(P〈0.05),but the levels of CGRP in the plasma and hippocampus of model group significantly decreased(P〈0.05).The levels of ET-1 and CGRP in the plasma and hippocampus of PNS low group and high dose group were significantly different(P〈0.05).The levels of ET-1and CGRP in the plasma and hippocampus of PNS high dose group vs PNS low dose group were significantly different(P〈0.05).PNS could relieve the deterioration of brain cell morphology,meainwhile it could decrease the death amounts of neurons in hippocampus after rats with brain trauma.Conclusion PNS can significantly decrease the levels of ET-1,increase the levels of CGRP in the plasma and hippocampus and relieve secondly brain damage after brain trauma.
