染色体9p21区域rs10757274多态性与缺血性脑卒中相关性的Meta分析 被引量:1

Association between rs10757274 in chromosome 9p21 and ischemic stroke:A comprehensive Meta-analysis

摘要 目的应用Meta分析方法探讨染色体9p21单核苷酸多态性(SNP)位点rs10757274与缺血性脑卒中(IS)易感性的关系。方法广泛检索中英文数据库中的相关研究,对符合纳入标准的每个研究采用纽卡斯尔-渥太华量表(NOS)进行质量评估。运用随机或固定效应模型计算比值比(OR),使用Q统计量检验法及I2统计量检测各研究之间的异质性,漏斗图和Egger检验评估发表偏倚。结果共纳入9篇文献10个研究,累计病例6746例,对照12577例。Meta分析结果显示,rs10757274基因多态性G等位基因增加IS的发病风险(OR=1.11,95%CI:1.06-1.16,P〈0.001);G等位基因或GG基因型增加了亚洲人大动脉粥样硬化脑梗死(LAS)的发病风险(等位基因模型:OR=1.18,95%CI:1.06-1.31,P=0.002;隐性模型:OR=1.33,95%CI:1.12-1.57,P=0.001)。未见发表偏倚。结论染色体9p21区域SNP rs10757274基因多态性与IS易感性相关。但该结论仍需大样本研究进一步验证。 Objective This study aimed to investigate the association between rs10757274 polymorphism in chromosome 9p21 and the susceptibility to ischemic stroke(IS)by using Meta-analysis;Methods Relevant studies were identified by searching databases extensively.The quality of included studies was assessed independently using the Newcastle-Ottawa Scale(NOS).Allele and genotype frequencies for each included study were extracted.The odds ratio(OR)was calculated using a random-effects or a fixed-effects model.The Q statistic and the I2 metrics were used to evaluate heterogeneity,and a subgroup analyses was also employed to explore possible specific susceptible genes.Begg′s test and Egger′s test were used to assess publication bias;Results A total of 10 studies within 9articles were included,which involved 6746 cases and 12577 controls.The results of Meta-analysis showed that the G allele of the rs10757274 polymorphism in chromosome 9p21 has an increased susceptibility to IS(OR=1.11,95%CI:1.06-1.16,P〈0.001).The G allele or GG genotype of the rs10757274 polymorphism may increase risk of large-artery atherosclerosis stroke among Asians(Allele contrast:OR=1.18,95%CI:1.06-1.31,P=0.002;Recessive model:OR=1.33,95%CI:1.12-1.57,P=0.001).There was no obvious publication bias.Conclusion Accumulated evidence so far indicated that the rs10757274 polymorphism in chromosome 9p21 is associated with the susceptibility of IS.Future studies with larger sample sizes will be necessary to confirm the present result.
