摘要
目的测定全蝎酶解产物中的蛋白质、多糖等成分的含量,并研究全蝎酶解产物的抗肿瘤活性。方法采用福林酚法和硫酸-苯酚法分别测定蛋白质和多糖的含量,MTT法测定不同浓度的全蝎酶解产物对肺腺癌细胞(A549)、乳腺癌细胞(MCF-7)、食管癌细胞(EC109)的抑制率。结果全蝎酶解产物中蛋白质含量为42.03%,多糖含量为13.84%;全蝎酶解产物对A549的抑制作用较强,且作用呈剂量依赖关系。结论全蝎酶解产物中蛋白质为主要成分,且抗肿瘤作用具有选择性。
Objective To determine the contents of protein and polysaccharide of the enzymolysis product of scorpion, and make research on its anti-tumor activity. Methods Lowry method and phenol-sulfuric acid method were used to determine the contents of protein and polysaccharide, respectively. The inhibition rates on lung adenocarcinoma cell line (A549), human breast cancer cell line (MCF-7) and esophageal cancer cell line (EC109) were determined using MTT assay. Results The contents of protein and polysaccharide of enzymolysis product of scorpion were 42.03 % and 13.84 %, respectively. The enzymolysis product of scorpion showed a better inhibitive effect on A549 in a dose- dependent manner. Conclusion The main component in enzymolysis products of scorpion is protein. The enzymolysis product of scorpion shows selective inhibitive effect on tumor cells.
