摘要
目的:从分子学水平检测腮腺沃辛瘤相对于瘤旁腮腺组织中生长抑制和DNA损伤修复基因45β( Gadd45β) mRNA表达情况并探讨其在沃辛瘤的形成及发展中的意义。方法搜集30例腮腺沃辛瘤组织及瘤旁正常腮腺组织。提取总RNA,然后进行逆转录得到cDNA,再进行实时荧光定量PCR检测,测出CT值,通过2-△△CT的相对定量的方法对荧光定量PCR检测的CT值进行标准化,从而比较肿瘤组织及瘤旁正常腮腺组织中Gadd45βmRNA表达情况。结果 Gadd45β在正常腮腺组织及肿瘤组织中均有表达,分别为1.516±0.104和1.177±0.089, Gadd45β在沃辛瘤中表达明显下调(P<0.05)。结论 Gadd45β表达下调可作为沃辛瘤发病机理一部分。
Objective To investigate the expression of growth arrest DNA damage?inducible 45β ( Gadd45β) mRNA in Warthin tumor of the parotid gland relative to normal parotid gland tis?sues and its significance in the development of Warthin tumor. Methods RNA was extracted from Warthin tumor and normal salivary gland tissues ( n=30 each) , reverse?transcripted to cDNA and subjected to real?time quantitative PCR. The CT value obtained was normalized by 2 -ΔΔCT method and the expression of Gadd45β mRNA was compared between tumor tissues and adjacent normal tis?sues. Results The expression level of Gadd45βmRNA in tumor tissues was 1. 177 ± 0. 089 , signif?icantly lower than that in normal tissues (1. 516 ± 0. 104, P〈0. 05) . Conclusion The down?ex?pression of Gadd45β gene may be involved in the development of Warthin tumors.
