缺血性脑卒中急性期血清脑特异性miRNAs水平的变化及与炎症应答和梗死体积的相关性 被引量:7

摘要 目的探讨缺血性脑卒中急性期血清脑特异性miRNAs水平的变化及其与炎症应答和梗死体积的相关性。方法收集起病24h内的缺血性脑卒中患者22例及健康对照8例,收集临床资料,NIHSS评分评估神经功能缺损,MRI检查评估梗死体积。于卒中起病24h内(22例)及48h内(6例)采取静脉血,分离血清。结果卒中起病24h内血清miRNA-124水平显著下降,起病48h内血清miRNA-219水平较起病24h内有所升高。卒中起病24h内血清miRNA-124、miRNA-9、miRNA-219水平均与梗死体积呈负相关,血清miRNA-9水平与入院时及入院24hNIHSS评分呈负相关。结论脑特异性miRNAs参与调节缺血性脑卒中急性期炎症应答水平,血清miRNA-124、miRNA-9、miRNA-219水平降低加剧炎症反应及脑组织炎症损伤。 Objective To evaluate the changes of the levels of serum specific miRNAs at acute stage of cerebral ischemic stroke and its correlation with inflammatory response and infarction volume.Methods Clinical data from 22 consecutive patients with acute ischemic stroke within 24 hafter symptom onset and 8healthy controls were retrospectively enrolled.Neurological function deficit was assessed by the National Institute of Health Stroke Scale(NIHSS)and infarction volume was evaluated by MRI.Venous blood was collected and serum was isolated from each patient within 24h(22cases)and 48h(6cases)after stroke onset.The levels of serum miRNA were detected by RT-PCR and the level of MMP-9by ELISA.Results The levels of serum miRNA-124 within 24hafter stroke onset were significantly decreased.The level of serum miRNA-219 within 48hafter stroke significantly rose up compared to that within 24 h.The levels of all the three miRNAs(miRNA-124,miRNA-9,miRNA-219)detected within 24 hafter stroke onset were negatively correlated with infarction volume.Additionally,the level of serum miRNA-9had negative correlation with NIHSS scores at admission and at 24 hours.Conclusion Brain specific miRNAs are involved in the regulation of inflammatory response in acute ischemic stroke.The lower levels of serum miRNA-124,miRNA-9and miRNA-219 can aggravate inflammatory response and exacerbate cerebral injury.
