摘要
本文研究了培养液中添加0、0.5、1.0、1.5和2.0 mg/m L NaHCO_3对球等鞭金藻(Isochrysis galbana)生长、细胞蛋白质含量及脂肪酸组成的影响,并进一步探讨了添加适宜NaHCO_3后不同培养阶段对细胞营养组成的影响。结果表明:NaHCO_3对球等鞭金藻的生长和细胞营养价值有显著影响。批次培养模式下培养液中添加1.5 mg/m L NaHCO_3可以获得最大细胞密度和比生长率。球等边金藻细胞中蛋白质含量随培养液中NaHCO_3添加量的增大而显著减少;而细胞中得单不饱和脂肪酸(MUFA)显著增加,多不饱和脂肪酸(PUFA)显著降低。在添加1.5 mg/m L NaHCO_3的培养条件下,不同培养阶段的藻细胞蛋白和脂肪酸组成也有显著变化,随着培养时间的延长,SFA和MUFA含量逐渐增加,PUFA含量则显著减少。培养液中添加NaHCO_3有助于提升球等鞭金藻作为生物柴油原料的品质。
The growth, cellular protein content and fatty acid profile in micro-algae Isochrysis galbana were valuated when the culture was supplemented with 0, 0.5, 1.0, 1.5 and 2.0 mg/m L NaHCO3 respectively. Then the protein content and fatty acid profile in cell at different growth phase were also investigated when the culture medium was added with 1.5 mg/m L NaHCO3. The results indicated there were significant effects of NaHCO3 supplementation on growth, cellular protein content and fatty acid profiles. The treatment supplemented with 1.5 mg/m L NaHCO3 displayed the highest cell density and special growth rate. The cellular protein content decreased significantly along with the increasing supplementation of NaHCO3 in culture medium. The monounsaturated fatty acids increased significantly while the polyunsaturated fatty acids decreased significantly along with the increase of NaHCO3 supplementation. There were significant differences in cellular protein content and fatty acid profile among the cells which were at a different growth phase when the cells were cultured in the medium supplemented with 1.5 mg/m L NaHCO3. The saturated fatty acids and monounsaturated fatty acids increased significantly while the polyunsaturated fatty acids decreased significantly along with the culture duration. Therefore it was suggested that NaHCO3 supplementation in culture medium was helpful for improving the micro-algae quality for bio-diesel production.
