泰山白皮松花粉对雏鸡免疫增强作用的初步研究

摘要 本试验设计旨在研究泰山白皮松花粉对雏鸡免疫功能的增强作用。取1日龄海兰褐公雏1050只,随机分为7组,3个松花粉佐剂组(Ⅰ、Ⅱ、Ⅲ),2个松花粉投喂组(Ⅳ、Ⅴ),另外2组分别为无松花粉对照组(Ⅵ)和正常对照组(Ⅶ)。以不同含量的泰山白皮松花粉作为免疫佐剂配制ND疫苗,胸肌注射接种各试验组雏鸡,9日龄1免,32日龄2免。Ⅰ、Ⅱ、Ⅲ、IV、V、Ⅵ组松花粉佐剂剂量分别为162μg/只,108μg/只,54μg/只,0,0,0;IV、V组6?11日龄投喂松花粉,剂量分别为0.3 g/只,0.1 g/只;VII组注射灭菌生理盐水。各试验组分别于1免后的第3d、7 d、14 d、21 d及2免后的第3 d、7 d、14 d、21 d采取血液,用β-微量法检测血清抗体。Ⅰ、Ⅱ、Ⅲ、Ⅵ组2免14 d后,每组随机取30只鸡采血进行体外淋巴细胞转化率检测。数据采用SPSS11.2软件进行T值检验分析。结果表明,Ⅰ、Ⅱ、Ⅲ组各指标均显著高于Ⅵ组(P<0.05),其中Ⅰ、Ⅱ组与Ⅵ组相比差异极显著(P<0.01)。Ⅳ、Ⅴ组显著高于Ⅵ组(P<0.05),Ⅵ组、Ⅶ组差异不显著。并且1免后14 d,2免后14 d、21 d的各试验组显著高于相应对照组(P<0.05)。因此,泰山白皮松花粉能提高雏鸡的抗体水平、血液淋巴细胞转化率。 An experiment was conducted to study the effects of immune enhancement caused by the Taishan white bark pine pollen on chicks. One thousand and fifty one-day-old Lyline male chicks were randomly divided into 7 groups: 3 pine pollen adjuvant groups(Ⅰ, Ⅱ, Ⅲ), 2 pine pollen feeding groups(Ⅳ,Ⅴ), the non-pine pollen control group(Ⅵ) and the control group(Ⅶ). The ND-vaccines were made by the immune adjuvant with different content of Taishan white bark pine pollen.The experimental group chicks were injected with the ND-vaccine in chest muscle when they were 9 and 32 days old, pine pollen does of groupⅠ, Ⅱ, Ⅲ, IV and V was 162 μg/feather, 108 μg/feather, 54 μg/feather, 0, 0, 0 respectively. Pine pollen was fed to group IV, V with 0.3 g, 0.1 g respectively when they were 6 and 11 days old. Group VII was injected with physiological saline. Samples of blood were taken on the 3rd, 7th, 14 th and 21 st day after each immunization. The lever of systemic antibody was detected by β-Microtest HA and HI. 30 chicks were randomly selected fromⅠ, Ⅱ, Ⅲ, Ⅵ group,Samples of blood were taken on 14 th day after the second immunization, the conversion rate of Lymphocytes was detected by MTT. SPSS11.2 software was used for T-score test analysis. The results showed that the indices in Group I, Ⅱ, Ⅲ were significantly higher than Group Ⅵ(P〈0.05). The differences between Group I, Ⅱ and Group Ⅵ were very significant(P〈0.01). Group Ⅳ,Ⅴ were significantly higher than Group Ⅶ(P〈0.05).The difference between Group Ⅵ and Ⅶ was not significant.And the test results of the groups on the 14 th day after first immunity,14 th and 21 st day after second immunization were significantly higher than that on the 3rd and 7th day(P〈0.05). Therefore Taishan white bark pine pollen could improve antibody level and lymphocyte transformation rate of chick.
