摘要
通过测定苯甲醛在279 nm处的吸光度来确定丝氨酸羟甲基转移酶的活性,利用单因素试验分析Escherichia coli SRZ018菌株最适产丝氨酸羟甲基转移酶的条件。结果表明,最适产酶条件为p H 8.0,反应温度50℃,CTAB为0.03 g/L,DL-β-苯基丝氨酸浓度为200μmol/L,PLP浓度为50μmol/L,该条件下丝氨酸羟甲基转移酶的活性最高达到0.881 U/mg。
The serine hydroxymethyl transferase activity was detected by determining benzaldehyde absorbance at 279 nm. The optimal conditions were proposed for improving the activity of serine hydroxymethyl transferase which was produced by E.coliSRZ018 strains by single factor experiment. The results showed that optimum enzyme production condition was p H 8.0,the optimal temperature 50 ℃,the concentration of CTAB,DL-β-phenyl serine and PLP were 0.03 g / L,200 μmol / L and 50 μmol / L,respectively,the serine hydroxymethyl transferase activity reached up to 0.881 U / mg.
