山楂叶总黄酮对2型糖尿病大鼠睾丸组织病变的改善作用 被引量:4

摘要 目的研究山楂叶总黄酮(HLF)对2型糖尿病大鼠睾丸组织病变的改善作用。方法将80只大鼠随机分为正常组、模型组和HLF低(50 mg/kg)、中(100 mg/kg)、高(200 mg/kg)剂量组,每组16只;除正常组外,其他各组均采用高脂高糖饮食8周后腹腔注射链脲佐菌素(STZ)的方法制备2型糖尿病大鼠模型;之后正常组、模型组灌胃生理盐水,HLF各组灌胃给予相应药物,每天1次,连续6周。干预结束后,测定各组大鼠空腹血糖水平,称量体质量并计算睾丸指数,测定血清睾酮以及睾丸组织中酸性磷酸酶(ACP)、乳酸脱氢酶(LDH)和谷氨酰转肽酶(γ-GT)含量;HE染色观察睾丸组织形态结构改变,透射电镜观察睾丸组织超微结构改变。结果与模型组比较,HLF中、高剂量组大鼠空腹血糖水平均显著降低,体质量及睾丸指数均显著升高,血清睾酮以及睾丸组织中ACP、LDH、γ-GT水平均显著上升,差异均具有统计学意义(P均<0.05)。HE染色后光镜下可见模型组大鼠睾丸曲细精管萎缩、生精细胞脱落且数量显著减少等病理改变,而HLF各组睾丸组织病变呈不同程度改善,以HLF高剂量组效果最显著;电镜下可见模型组睾丸组织胞体缩小、细胞器减少、内质网扩张、脂滴形成、细胞核固缩、等病理改变,而HLF各组睾丸细胞超微结构病变明显改善,以HLF高剂量组最为显著。结论 HLF对2型糖尿病大鼠睾丸组织病变具有一定的改善作用。 Objective It is to investigate the ameliorate effects of Hawthorn Leaves Flavonoids( HLF) on the testicular tissue lesion in type 2 diabetic mellitus( T2DM) rats. Methods Eighty rats were randomly divided into five groups: normal group,model group and HLF low( 50 mg / kg),medium( 100 mg / kg),high( 200 mg / kg) dose group,16 cases in each group. All the rats were made into T2 DM models by high- fat diet and injected STZ except the normal group; the drugs or normal saline were given by intragastric administration for 6 weeks,once a day. After the intervention,the level of plasma glucose was determined,the body weight and testis tissue index was calculated,the content of testosterone in serum and ACP,LDH,γ- GT in testis tissue were detected; the histopathological changes of testis tissue was observed by HE staining,the ultrastructure of testis tissue was observed by electron microscope. Results Compared with model group,the level of plasma glucose were significantly decreased,the body weight,the testis tissue index and the content of testosterone were significantly increased,the content of ACP,LDH,γ- GT in testis tissue were significantly increased in HLF high and medium dose groups,there were significant difference( all P 0. 05). After HE staining under light microscope,the testicular seminiferous was atrophied,germ cell was ablated and the number was significantly reduced in model group; however,the testicular tissue lesions were improved in different degrees in all the HLF groups,and the effect of HLF high dose group was the most significant. Under the electron microscope,the cell body of testicular tissue was significantly reduced,and organelle was reduced,endoplasmic reticulum was expanded,and lipid droplet was formed,and nuclear was condensed in model group; however,the ultra-structural changes of testicular cells were significantly improved in all the HLF groups,and those of HLF high dose group was the most significant. Conclusion HLF has a protective effect on the testicular tissue of rats with T2 DM.
