摘要
目的筛选制备兔富血小板纤维蛋白(PRF)和改良型富血小板纤维蛋白(A-PRF)凝胶的方法 ,为后续动物实验及临床研究提供参考。方法选择17种离心方法制备兔PRF凝胶,16种离心方法制备兔A-PRF凝胶,通过光镜观察,与人标准方法制备的PRF、A-PRF对比分析,筛选与人PRF、A-PRF镜下微观结构最接近的兔PRF、A-PRF制备方法。结果各组离心产物表现不一,凝块结构分层、活细胞数量范围、纤维蛋白排列各不相同。其中3250 r/min离心10 min制备的兔PRF,2000 r/min离心24 min制备的兔A-PRF分别与人PRF、A-PRF镜下微观组织学结构最为接近。结论 3250 r/min离心10 min和2000 r/min离心24 min分别可作为制备兔PRF和A-PRF的最佳条件。
Objective To screen the methods for preparing rabbit platelet-rich fibrin(PRF) and advanced platelet-rich fibrin(APRF) gel and provide reference for animal experiment and clinical research in the future.Methods A total of 17 centrifugal methods for preparing rabbit PRF gel and 16 centrifugal methods for preparing rabbit A-PRF gel were selected.Such gel was compared,via light microscope,with the human PRF and A-PRF gel prepared by standard method to screen the method by which rabbit PRF and A-PRF gel with closest microscopic structure to that of human PRF and A-PRF gel.Results The products of the groups were different.The structure stratification of the clot,the number of living cells and the arrangement of the fiber protein were different,too.The microscopic structure of rabbit PRF prepared by 3250 r/min for 10 min and rabbit A-PRF by 2,000 r/min for 24 min was the closest to that of human PRF and A-PRF respectively.Conclusion 3250 r/min for 10 min and 2000 r/min for 24 min may be the best conditions for preparing PRF and A-PRF.
