七氟醚后处理对大鼠心肌缺血再灌注氧化应激的影响 被引量:7

摘要 目的探讨七氟醚后处理对体内大鼠心肌缺血再灌注损伤时氧化应激的影响。方法选择健康雄性成年SD大鼠64只,随机分为假手术组、单纯七氟醚组、缺血再灌注组和七氟醚后处理组,每组16只。再灌注末,利用动物超声仪测定心功能指标,采用1%TTC测定心肌梗死面积百分比,并检测蛋白羰基化水平、心肌还原型谷胱甘肽(GSH)、氧化型谷胱甘肽(GSSG)和丙二醛含量以及心肌超氧化物歧化酶2(SOD2)和血红素氧合酶1(HO-1)蛋白表达水平。结果与假手术组比较,缺血再灌注组心功能指标恶化,心肌梗死面积百分比、蛋白羰基化、丙二醛、SOD2和HO-1蛋白表达明显上调,心肌GSH和GSH/GSSG明显降低[(6.43±0.70)nmol/L vs(15.94±2.20)nmol/L,2.04±0.28 vs 4.82±0.50,P<0.05]。与缺血再灌注组比较,七氟醚后处理组心功能指标改善,心肌梗死面积百分比、蛋白羰基化、丙二醛明显降低,心肌GSH、GSH/GSSG、SOD2和HO-1蛋白表达明显上调(P<0.05)。结论七氟醚后处理可减少体内大鼠心肌缺血再灌注损伤,发挥心肌保护作用,且氧化应激反应是其心肌保护作用的重要机制之一。 Objective To study the effect of sevoflurane postconditioning on oxidative stress in rats following myocardial I/R injury.Methods Sixty-four healthy male adult SD rats were randomly divided into sham operation group,simple sevoflurane group,I/R group,and sevoflurane postconditioning group(16in each group).Their cardiac function was detected by echocardiography,their myocardial infarct size was measured by 1% TTC,and expressions of protein carbonyl,GSH,GSSG,MDA,SOD2 and HO-1protein were detected at the end of reperfusion.Results The cardiac function was poorer,the myocardial infarct size was significantly larger,the expression levels of protein carbonyl,MDA,SOD2 and HO-1protein were significantly higher whereas the GSH expression level and the GSH/GSSG ratio were significantly lower in I/R group than in sham operation group(P〈0.05).The cardiac function was better,the myocardial infarct size was significantly smaller,the expression level of protein carbonyl was significantly lower whereas the expression levels of GSH,SOD2 and HO-1protein and the GSH/GSSG ratio were significantly higher in sevoflurane postconditioning group than in I/R group(P〈0.05).Conclusion Sevoflurane postconditioning can alleviate myocardial I/R injury in rats and protect their myocardium with oxidative stress response as one of its important mechanisms.
