摘要
目的 采用高效液相色谱法测定翻白叶树中原儿茶酸的含量。方法 采用Phenomenex ODS2色谱柱(250 mm×4.6mm,5μm),流动相为乙腈-0.1%冰乙酸溶液(5∶95),流速1.0 m L·min-1,检测波长260 nm,柱温30℃。结果 原儿茶酸0.0155~0.3100μg与峰面积呈良好的线性关系(r=0.9994);平均加样回收率为98.71%,RSD=2.98%(n=6)。结论 所用方法简单便捷、灵敏度高、重复性好。
OBJECTIVE To establish an HPLC method for determining protocatechuic acid in Pterospermum heterophyllum Hance.METHODS The method was used with the chromatographic column of Phenomenex ODS2(250 mm × 4.6 mm),and a mixture of acetonitrile-0.1% glacial acetic acid(5 ∶95) was used as the mobile phase at a flow rate of 1.0 m L·min-1.The UV detection wavelength was 260 nm and the column temperature was 30 ℃.RESULTS The calibration curves of protocatechuic acid was in good linearity over the range of 0.0155-0.3100 μg(r = 0.9994) and the average recovery of protocatechuic acid was 98.71% with the RSD of 2.98%(n = 6).CONCLUSION The method is simple,sensitive and accurate with a high reproducibility.
