摘要
目的检测KISS1在人肺腺癌组织、癌旁组织中的表达情况,分析两者表达差异,探讨KISS1在人肺腺癌发生中的作用。方法利用免疫组织化学染色、免疫印迹法(Western blot)及实时定量PCR(real-time PCR)检测28例人肺腺癌组织及对应癌旁组织中KISS1蛋白及m RNA的表达,应用卡方检验对结果进行分析。结果免疫组化及Western blot结果显示KISS1蛋白在癌组织中表达水平显著低于癌旁组织。RT-PCR结果显示在肿瘤组织中KISS1m RNA表达的平均倍数显著低于癌旁组织(0.26±0.10 vs 1.02±0.13,<0.01)。结论 KISS1在肺腺癌表达水平低于癌旁正常组织。
Objective To detect the expression of KISS1 in lung adenocarcinoma and the paracancer normal tissues to analysis the role of KISS1 in the carcinogenesis of lung adenocarcinoma. Methods The expression of KISS1 protein and m RNA in 28 cases of lung adenocarcinoma and the paracancer normal tissues was detected by immunohistochemical staining, Western blot and Real-time PCR respectively. The results were analyzed by chi square test. Results Immunohistochemistry and Western blot showed that the expression level of KISS1 protein was significantly lower in cancer tissue than in paracancer normal tissues. RT-PCR showed that the expression level of KISS1 m RNA was significantly lower in cancer tissue than in paracancer normal tissues( 〈0.01). Conclusion The expression level of KISS1 was lower in lung adenocarcinoma than in paracancer normal tissues.
