摘要
目的研究新型抗癌药物麦普替林对人源黑色素细胞瘤A375细胞的作用,探讨其对黑色素细胞瘤治疗效果。方法应用不同浓度的麦普替林药物对A375细胞进行处理,比色定量法检测细胞的凋亡水平。在A375细胞中分别加入无菌水和麦普替林,进行划痕实验;利用Transwell实验,研究麦普替林对于黑色素瘤转移和侵袭的影响。结果麦普替林作用下A375细胞凋亡水平显著增加。划痕实验和Transwell实验显示,A375细胞加入麦普替林后,其增殖、迁移、侵袭的细胞数量都有显著的减少。结论麦普替林显著抑制黑色素瘤,具有良好临床应用前景。
Objective To study the therapeutic effect of maprotiline on human melanoma cell line A375. Methods The apoptosis of A375 cells was evaluated by colour matix, the migration and invation of A375 cells were examined by wound healing test and transwell. Results Maptotiline induced the apoptosis of A375 cells significantly, inhibited the migration and invasion as well as proliferation of A375 cells demonstrated by wound healing and transwell. Conclusion The suppressing effect of maprotiline on melanoma A375 cells suggests that it would be a promising chemotherapeutic drug against melanoma.
