摘要
目的探讨β-七叶皂甙钠对脑外伤小鼠脑皮质水通道蛋白4(AQP4)与水通道蛋白5(AQP5)表达的影响。方法昆明小鼠120只,随机分为假手术组(40只)、脑外伤组(40只)和β-七叶皂甙钠组(40只)。采用Feeney自由落体法制作脑外伤模型,采用干湿重法测定脑含水量,免疫组化和Western blot方法检测各组小鼠大脑皮质AQP4与AQP5的表达。结果相比于对照组,脑外伤后3h,小鼠脑含水量明显上升,6h继续升高,24h达峰值,3d时开始下降;相比于脑外伤组,相同时间点的β-七叶皂甙钠组小鼠脑含水量则明显降低(<0.05)。相比于对照组,脑外伤后3h,大脑皮质AQP4与AQP5表达水平明显增高,24 h后略有降低,3d时再次明显增高,并达峰值;相比于脑外伤组,相同时间点的β-七叶皂甙钠组小鼠大脑皮质及海马AQP4与AQP5的含量则明显降低(<0.05)。结论β-七叶皂甙钠参与脑外伤水肿的调节可能与下调AQP4与AQP5蛋白表达相关。
Objective To study the effect of β-sodium aescinate on the expressions of aquaporin-4 and aquaporin-5 in the cortex of mice after traumatic brain injury. Methods 120 Kunming mice were randomly divided into:sham group, traumatic brain injury group and β-sodium aescinate group averagely. The traumatic brain injury model was established according to Feeney’s method.Dry-wet weight method were used to measure the extent of brain edema,the expression of aquaporin-4 and aquaporin-5 in the cortex was detected by immunohistochemistry and western blot. Results The brain water content was increased significantly at 3h, reached peak at 24 h, began to drop at 3d in traumatic brain injury group than in the sham, but decreased significantly in β-sodium aescinate group than in traumatic brain injury group. The expression levels of aquaporin-4 and aquaporin-5 protein in cortex were increased significantly at 3h, reached peak at 3d in traumatic brain injury group than in sham, but decreased significantly in β-sodium aescinate group than in traumatic brain injury group. Conclusion The regulatory effect of β-sodium aescinateeon dema of traumatic brain injury might be related to the downregulation of AQP4 and AQP5 expressions.
