摘要
目的观察抗肿瘤中药半枝莲不同极性提取物对人胚胎肾脏细胞体外活性的影响。方法分别提取半枝莲的水、乙醇、甲醇、乙酸乙酯和正丁醇等5种提取物,用高效液相色谱(high performance liquid chromatography,HPLC)检测不同极性提取物的成分差异,用MTT法测定半枝莲不同极性提取物对人胚胎肾脏细胞HEK293增殖的影响。结果半枝莲不同极性提取物组成呈现明显差异,乙醇提取物与甲醇提取物的组成较为相似,而正丁醇提取物与乙酸乙酯提取物较为相似,水提取物则与其他提取物均存在较大差异。细胞活性试验表明,水、乙酸乙酯和正丁醇提取物(0.01、0.1、1、20 g/L)培养48小时后对HEK293的体外增殖无明显影响,而用浓度为0.01、0.1、1、20g/L的甲醇提取物和0.1、1、20 g/L的乙醇提取物培养48小时后对细胞增殖则呈现一定程度的抑制(均P<0.05)。结论本研究提示,抗肿瘤中药半枝莲的水、乙酸乙酯和正丁醇提取物对肾细胞活性无影响,而甲醇和乙醇提取物存在细胞活性抑制现象。
Objective To study the influence of different polarity extract from Scutellaria barbata( S. barbata) on the viability of human embryonic kidney 293( HEK293) cells. Methods S. barbata extracts were extracted by water,ethanol,methanol,butanol,and acetic ether,respectively. High performance liquid chromatography( HPLC) was used to scan the fingerprint of the extracts of different polarities. The MTT method was used to test the influence of different polarity extracts on the proliferation of HEK293 cells. Results The components from different polarity extracts of S. barbata showed significant difference. Ethanol and methanol extracts were relatively similar in composition. Butanol and acetic ether extracts also showed high similarity. However,the water extracts were significantly different with the other extracts. Moreover,cell viability tests suggested that only the ethanol( 0. 1,1,20 g / L) and methanol( 0. 01,0. 1,1,20 g / L) extracts were able to inhibit the proliferation of HEK293 cells( P〈0. 05),whereas the other three extracts had no obvious effect. Conclusion The ethanol and methanol extracts from S. barbata are able to inhibit the in vitro viability of HEK293 cells,thus one should be cautious of their renal toxicity in clinical application.
