摘要
目的研究微小RNA在宫颈癌的早期筛查中的应用及预后价值。方法采用实时定量PCR(定量PCR)测定宫颈癌肿瘤组织中miRNA的表达水平(miR-21,miR-27a,miR-34a,miR-155,miR-196a,miR-203)。结果 miR-27a在宫颈上皮内瘤变(CIN2-3)中的表达水平显著高于CIN1(P=0.023),而在鳞状细胞癌(SCC)中的表达水平高于CIN2-3(P=0.033)。此外,miR-34a在CIN2-3中的表达水平显著低于CIN1(P=0.041),在SCC中的表达水平低于CIN2-3(P=0.021)。与CIN1比较,CIN2-3的miR-27a表达水平上调(P=0.028)而miR-34a表达水平下调,HPV16阳性与宫颈癌显著相关(CIN2-3/CIN1:P=0.027;SCC/CIN2-3:P=0.036)。miR-34a的表达水平与吸烟状况和HPV16相关。结论检测miRNA表达水平有助于评估不同宫颈癌变和预测HPV感染结果。
Objective To study the application and prognostic value of small RNA in early screening of cervical cancer.Methods Real-time quantitative PCR( quantitative PCR) was used to determine the expression levels of miRNA cervical cancer tissues( miR-21,miR-27 a,miR-34 a,miR-155,miR-196 a,miR-203). Results miR-27 a expression level in cervical intraepithelial neoplasia( CIN) 2-3 was significantly higher than CIN1( P = 0. 023),while the expression levels of squamous cell carcinoma( SCC) was higher than CIN2-3( P = 0. 033). In addition,miR-34 a expression level in CIN2-3 was significantly lower than CIN1( P = 0. 041),and in SCC was lower than CIN2-3( P = 0. 021). Compared with CIN1,miR-27 a expression of CIN2-3 increased( P = 0. 028) and miR-34 a decreased,HPV 16 positive expression was correlated with CIN( CIN2-3 /CIN1: P = 0. 027; SCC/CIN2-3: P = 0. 036). miR-34 a expression level and smoking status and HPV16 were correlated. Conclusion The detection of miRNA expression level can help evaluate cervical lesions and HPV infection.
