当归补血汤作用于四膜虫抗氧化基因——抗氧化药物筛选新方法初探

摘要 目的:以经典方当归补血汤对四膜虫生长曲线和抗氧化酶基因的作用探讨筛选抗氧化药物的新方法。方法:将不同剂量的当归补血汤接种于嗜热四膜虫的培养液中,以无菌蒸馏水为空白对照,观察嗜热四膜虫的生长曲线,另外,对上述不同培养液中的嗜热四膜虫采用Real Time-PCR法测定嗜热四膜虫体内的谷胱甘肽过氧化物酶(glutathione peroxidase,GPx4)、硫氧还蛋白还原酶(Thioredoxin reductases,TxnRd2)的基因表达。结果:当归补血汤干预组生长曲线与空白组相比有明显的提高。与空白组相比,嗜热四膜虫的GPx4、TxnRd2基因表达均显著升高。结论:当归补血汤能够明显改善四膜虫生长曲线,促进GPx4、TxnRd2基因mRNA表达,提示生物体生长曲线可能与抗氧化酶基因表达有关。 Objective:To study the effects of Danggui Buxue decoction on mRNA expression of GPx4 and TxnRd2 in Tetrahymena thermophile.Methods:T.thermophila growth was intervened by using the boil-free granule of traditional Chinese medicine.Four different concentrations of Danggui Buxue decoction were added into culture medium of T.thermophile,and each concentrations with two parallel designs,by setting a group using sterile distilled water as blank control.Processing of gene glutathione peroxidase (GPx4) and thioredoxin reductases (TxnRd2) expression data generated by quantitative real-time RT-PCR.Results:Compared with blank control group,the growth curve of medicine intervention went up significantly.Results also showed that the expression level of GPx4 and TxnRd2 were increased by Danggui Buxue decoction.Conclusion:Danggui Buxue decoction could increase the T.thermophile’s growth curve,which may be related to GPx4 TxnRd2 gene expression.
