摘要
目的运用基因芯片技术检测针刺泻法对应激性高血压前期大鼠肾脏基因表达的影响并分析其内在机理。方法雄性Wistar大鼠27只,随机分为空白组、模型组、模型+针刺泻法组,每组9只。造模方法:噪声结合足底电击刺激,周期为14d。造模同时针刺泻法组取曲池穴、太冲穴进行干预,模型组和空白组每天做与针刺组相同的抓取刺激。在造模前1天、第3、5、7、9、11、13、15天测量大鼠的血压,并观察其生理学行为学改变。第15天量取血压后取材,用Rat Gene 2.0 Array technology进行基因芯片分析。结果造模期间空白组血压始终保持正常水平。模型组在造模第3天即突破了120 mmHg(P〈0.01),并在后来的第5、7、9、11、13、15天持续升高至较高水平(P〈0.01)。造模过程中大鼠出现尖叫、咬架、小便变黄、毛质粗糙、眼睛充血等变化,且血压始终处于120~139 mmHg范围内,说明造模成功。模型+针刺泻法组的血压值在第5、7、9、11、13、15天均明显低于模型组。基因表达频谱结果显示:与模型组相比,模型+针刺泻法组有106个基因表达升高,92个基因表达降低。其中高血压相关基因CYP1A1、IGFBP-1的表达也发生相应变化。结论针刺太冲、曲池并施以泻法,可明显降低应激性高血压前期大鼠的血压,并影响其肾脏的基因表达。
Objective To study the effect of twirling-reducing needling manipulation( TRNM) on gene expression in kidney of stress-induced pre-hypertension rat by gene chip technology. Methods Twenty-seven,male Wistar rats were randomly divided into 3 groups( n = 9 / group) : blank control group,model control group and model + TRNM group. Foot-shocks combined with generated noise was applied in the preparation of stress-induced pre-hypertension model and lasted for 14 days. Twirling-reducing needling manipulation was used on Taichong( LR3) and Quchi( LI11)of TRNM group along the molding cycle. The model control and blank control group rats were grabbed in the same restrainers for 20 min without any other intervene. Systolic blood pressure( SBP) was tested one day before work and on the 3 rd,5 th,7 th,9 th,11 th,13 th and 15 th day. Gene expression profiles was determined by Rat Gene 2. 0 Array technology. Results SBP of blank control group rats stayed normal along the modeling cycle. At the 3rd day,SPB of model control group elevated to 120 mm Hg( P〈0.01),and continuously rose to 139 mm Hg,along with changes like irritation,squealing,yellow urination and bloodshot eyes,which indicates the success of modeling. SBP of TRNM group was significantly lower( P 〈0.01) than model control group since the 5 th day. Microarray analysis showed that compared with model control group,106 genes were upregulated and 92 genes were downregulated in TRNM group. The expression of CYP1A1 and IGFBP-1,which have a close relationship with hypertension and organ damage,was significantly changed by Twirling-reducing needling. Conclusion Twirling-reducing needling manipulation at Taichong and Quchi can significantly lower the SBP of stress ﹣ induced pre-hypertension rats and effect its gene expression in kidney.
