冻干重组人脑利钠肽对兔心肌梗死后心肌组织病理改变的影响及机制 被引量:12

摘要 目的观察冻干重组人脑利钠肽(rhBNP)对兔心肌梗死后心肌组织病理改变的影响,并探讨其作用机制。方法将32只新西兰白兔随机分为假手术组8只、模型组12只、rhBNP组12只。模型组、rhBNP组采用结扎冠脉前降支的方法制备心肌梗死模型,假手术组只开胸不结扎冠状动脉。记录梗死前后十二导联心电图变化,明确模型制备成功。rhBNP组术后即刻及术后每间隔12 h耳缘静脉推注rhBNP 7μg/kg,直至术后72 h。术后4周取兔心肌组织行HE染色,观察心肌细胞形态及炎症细胞浸润情况;采用Western blotting法检测各组心肌组织IL-1β、TGF-β1表达。结果模型组、rhBNP组梗死心肌细胞变形、肿胀、破裂、坏死,排列杂乱,周围炎症细胞浸润,rhBNP组上述改变程度均较模型组减轻。与假手术组比较,模型组、rhBNP组心肌组织IL-1β、TGF-β1表达均增加(P<0.05或<0.01);与模型组比较,rhBNP组心肌组织IL-1β、TGF-β1表达均降低(P均<0.01)。结论 rhBNP可减轻兔心肌梗死后的心肌组织病理损伤,抑制心肌组织炎症因子IL-1β、TGF-β1表达可能是其作用机制。
