摘要
目的通过对达托霉素产生菌进行诱变选育研究,以期获得达托霉素高产菌株。方法采用常压室温等离子体诱变育种技术,结合紫外诱变技术对达托霉素产生菌玫瑰孢链霉菌进行复合诱变,并以谷氨酸钠耐受为选择压力进行理性筛选。结果经诱变选育获得一株达托霉素产量达到3.9g/L的突变株Q12-63#,发酵单位较出发菌株提高了37%。结论常压室温等离子体及紫外复合诱变方式,结合谷氨酸钠耐受性筛选模型,达托霉素产生菌正突变几率高,可有效提高菌株的达托霉素产量。
Objective To obtain a high yield daptomycin-producing strain by means of mutation breeding. Methods Streptomyces roseosporus, daptomycin-producing strain was breeded by combination mutation of atmospheric and room temperature plasma (ARTP) and UV, with sodium glutamate as selection factor. Results A mutant strain named Q12-63# was obtained and the yield of daptomycin reached 3.9g/L which was 37% higher than that of the original one. Conclusion The combination mutation method of ARTP and UV, with sodium glutamate as selection factor for breeding has high positive mutation rate and the yield of daptomycin is significantly increased.
