摘要
目的研究探讨胡芦巴碱、五味子油和二甲双胍联合治疗对2型糖尿病大鼠糖脂代谢和海马区Neuritin表达的影响。方法采用链脲佐菌素+高糖、高脂饲料喂养,建立2型糖尿病大鼠模型。糖尿病大鼠分为糖尿病模型组、药物联合治疗和罗格列酮组,每组11只大鼠,连续给药8周。另取10只正常SD大鼠作为正常对照组。检测各组大鼠空腹血糖(FBG),糖化血红蛋白(HbA1c)、空腹血清胰岛素(FINS)、总胆固醇(TC)、三酰甘油(TG)、肝糖原和肌糖原水平,实时聚合酶链反应、Western blot检测分析海马区Neuritin表达水平。结果给药8周后,糖尿病模型组大鼠FBG、HbA1c、FINS、TC、TG升高,胰岛素敏感指数(ISI)、肝糖原和肌糖原水平、海马Neuritin mRNA及蛋白表达水平下降,与正常对照组比较差异有统计学意义(P<0.01)。与糖尿病模型组比较,药物联合治疗组和罗格列酮组大鼠FBG、HbA1c、FINS水平下降,而ISI、肝糖原和肌糖原水平、海马Neuritin mRNA及蛋白表达水平上升,差异有统计学意义(P<0.01);药物联合治疗组与糖尿病模型组比较,TC、TG下降,差异有统计学意义(P<0.01),而与罗格列酮组TC、TG比较,差异无统计学意义(P>0.05)。结论胡芦巴碱、五味子油和二甲双胍联合治疗能改善糖尿病大鼠糖脂代谢和海马区Neuritin表达。
Objective To observe the effect of trigonelline, Fructus Schisandrae oil and Metformin therapy on neuritin expression in hippocampal area and glucose and lipid metabolisms in rats with type 2 diabetes. Methods Type 2 diabetic rat model was established by feeding Streptozotocin plus high-sugar and high-lipid diet. The diabetic rats were divided into diabetic model group, combined drug treatment group and Rosiglitazone group with 11 rats in each group. The rats received continuous treatment for 8 weeks. The normal control group consisted of 10 normal SD rats. FBG, HbAlc, TC, FINS, TG, liver glycogen and muscle glycogen levels were tested. Real-time PCR and Western blot were used to determine neuritin expression level in hippocampus. Results Compared with the normal control group, the FBG, HbAlc, FINS, TC and TG levels were increased significantly; the insulin sensitivity index (ISI), liver glycogen and muscle glycogen levels, hippocampal neuritin mRNA and protein expressions were decreased significantly in the diabetic model group after treatment for 8 weeks (P〈0.01). Compared with the diabetic model group, the FBG, HbA1c and FINS levels were significantly decreased; but ISI, liver glycogen and muscle glycogen levels, and the expression levels of hippocampal neuritin mRNA and protein were significantly increased in the combined drug treatment group and Rosiglitazone group (P〈 0.01). Compared with the diabetic model group, TC and TG decreased significant- ly in the combined drug treatment group (P〈 0.01); but there were no significant differences in TC or TG between the diabetic model group and the Rosiglitazone group (P 〉 0.05). Conclusions Combined trigonelline, Fructus Schisandrae oil and Mefformin therapy can improve glucose and lipid metabolisms and expression of neuritin in the hippocampus of type 2 diabetic rats.
