冷应激对大鼠肝脏组织中Rap1b表达的影响 被引量:2

摘要 目的探讨冷应激对大鼠肝脏组织中Rap1b表达的影响。方法将20只Wistar大鼠随机分为常温饲养组和冷应激组,每组10只,正常饲养温度为(24.0±0.1)℃,冷应激温度为(4.0±0.1)℃。经12 h冷刺激后,采集大鼠肝脏组织,提取总RNA和总蛋白,通过实时荧光定量PCR和Western blot法分别检测Rap1b基因m RNA的转录水平和蛋白的表达水平。结果冷应激组大鼠肝脏组织中Rap1b基因m RNA的转录水平和蛋白表达水平均显著高于常温饲养组(P<0.01)。结论冷应激可显著提高大鼠肝脏组织中Rap1b基因m RNA的转录水平和蛋白表达水平。 Objective To investigate the effect of cold stress on Rap1 b expression in rat liver tissue. Methods Twenty Wistar rats were randomly divided into two groups, ten for each. The rats in room temperature raising group were raised at(24. 0 ± 0. 1)℃, while those in cold stress group at(4. 0 ± 0. 1)℃. The liver tissues of rats 12 h after cold stress were collected, from which the total RNA and total protein were extracted for determination of m RNA transcription and protein expression levels of Rap1 b by real-time fluorescent quantitative PCR and Western blot respectively. Results Both the m RNA transcription and protein expression levels in cold stress group were significantly higher than those in room temperature raising group(P〈 0. 01). Conclusion Cold stress enhanced the m RNA transcription and protein expression of Rap1 b in rat liver tissue significantly.
