脱水淫羊藿素促进骨髓间质干细胞修复裸鼠临界性颅骨缺损的实验研究 被引量:2

摘要 目的研究脱水淫羊藿素在体内对裸鼠临界性颅骨缺损修复的影响。方法体外培养并流式细胞仪鉴定人骨髓间质干细胞;建立裸鼠颅骨临界性骨缺损,用脱水淫羊藿素及脱水淫羊藿素复合骨髓间质干细胞修复骨缺损。修复2个月后,用micro-CT检测脱水淫羊藿素的骨缺损修复能力的影响。结果分离并培养了人骨髓间质干细胞,流式细胞仪证实人骨髓间质干细胞是一群CD73+CD90+CD34-CD45-细胞。Micro-CT结果显示,与空白对照组对比,脱水淫羊藿素组的新增骨量增加。而脱水淫羊藿素复合骨髓间质干细胞后可以显著提高骨缺损处的新骨形成量(P<0.01)。结论脱水淫羊藿素在体内具有促进骨缺损修复作用,而脱水淫羊藿素复合骨髓间质干细胞可以显著增强骨缺损修复能力。 Objective To investigate the effects of anhydroicaritin on the repair of the critical - sized cranial bony defect in nude mice. Methods Bone marrow mesenchymal stem cells (BMSCs) were cultured and identified by flow eytometry. Critical - sized cranial bone defect was created, then filled with alginate hydrogel (blank control group), anhydroicaritin in alginate hydrogel, or anhydroicaritin plus BMSCs in alginate hydroge. All animals were scarified at 2 months post - transplantation for micro - CT examination. Results The cultured BMSCs were characterized as CD73^+ CD90^+ CD34^- CD45^- cells. Micro - CT results showed that compared with blank control group, newly - formed bone was slightly but significantly increased in anhydroicaritin group, and markedly increased in anhydroicaritin plus BMSCs group ( P 〈 0.01 ). Conclusion Anhydroicaritin could promote the repair of bone defect in vivo, and the repair ability could be significantly en- hanced when anhydroicaritin is used in combination with BMSCs.
