大鼠前扣带回皮层吻侧部精确取样的方法

The Method of Taking the Rostral Anterior Cingulate Cortex Tissue Precisely from Rat Brain

摘要 目的:自制大鼠脑组织取样模具,精确切取双侧大鼠前扣带皮层吻侧部。方法:(1)自制动物脑组织精确取样模具;(2)在低温下将大鼠脑组织冠状切为脑组织片;(3)按照自制取材模具上的定位标识将已切好的大鼠脑片摆放好,逐步对组织段切割,最终可得到样本量较精确的大鼠前扣带皮层吻侧部样品。之后即可进行该脑区的蛋白、核酸等其他方面的研究。结果:实验中切取不同个体大鼠脑组织的部位和体积基本一致,不同动物个体取样差异小,实验结果可比性更强,且切取的组织大小不因操作者不同而异。结论:自制大鼠前扣带皮层吻侧部精确取样模具的使用简便易行,取样部位、大小一致性好,方法经济可靠,使后续实验结果更准确;该方法可推广应用于大鼠其他脑区的取样或其他小动物脑组织的取样。 Objective:To take the rostral anterior cingulate cortex(r ACC) tissue precisely from rat brain with home-made rat brain cutting mold.Method:(1)The cutting mold was made by ourselves.(2)The rat's brain was cut into coronary sections at low temperature.(3)After that,the tissue section was put on the homemade cutting mold according to the marked line,it might be cut step by step. We finally got accurate sample size at exact location from r ACC brain area in rat.After the above,the following studies on the detection of protein or nucleic acid may proceed.Result:In this way,we got accurate r ACC brain area sample tissues which were in the same size and location in rat, regardless of different individual experimental animals we used or each new experiment operator.Conclusion:Home-made cutting mold is easy to make and use, and it can be well applied to other brain area in rat or other small animals.
